Objective:Paridis rhizoma?RP?,the dried rhizome of Paris polyhoylla Smith var.yunnanensis?Franch.?Hand.-Mazz?PPY?and P.polyhoylla Smith var.chinensis?Franch.?Hara?PPC?widely used to treat antipyretic,analgesia,antidote and acute nephritis,and et al.PPC were extracted and fractionated in our past study,and the chemical compositions in the total extract and fractions were characterized.In order to further understand the chemical composition of PPC,and study the characteristics of the pharmacological effects and conclusion the structure-activity relationship,the present study was carried out by evaluating analgesic,anti-inflammatory and hemolysis effects of extract and fractions of PPC,and investigating the cytotoxic and anti-inflammatory activity of pure compounds.Besides,the pharmacological and hemolytic effect of extract and fractions of the aerial parts of PPCwere investigated for the development and utilization of aerial parts of PPC.Methods:Using hot plate experiment,Hot prick test,acetic acid body torsion experiment,the tail suspension test,Xylene induced mouse ear swelling model and red blood cells in vitro hemolysis test,Hep G2 cell growth inhibition test,cell damage experiment of RAW264.7 to evaluate the pharmacological activity of extractive,macroporous resin elution part and compounds from PPC.Results:The rhizome and aboveground part extracts of PPC all showed analgesic anti-inflammatory and hemolysis effects,three kinds of saponins from rhizome 70%parts have anti-inflammatory effects.The saponins include pennogenin have strongest hemolysis effect.13 compounds were isolated and 10 compounds'structures were determinedasfollows:3?-O-?-L-glucopyranosyl-?1?3?-[?-L-rhamnopyranosyl-?1?2?]-?-D-glucopyranosylhomo-aro-cholest-5-ene-26-O-?-D-glucopyranoside?6?,pregnane-5,16-diene-3?-alcohol-20-keto,3?-O-?-L-rhamnopyranosyl?1?2?[?-L-rham nopyranosyl?1?4?]-?-D-glucopyranoside?4?,3?-O-?-L-rhamnopyranosyl-?1?4?-O-?-L-rhamnopyranosyl-?1?4?-[O-?-L-rhamnopyranosyl-?1?2?]-O-?-D-glucopyranosyl homo-aro-cholest-5-ene-26-O-?-D-glucopyranoside?5?,3-O-?-L-arabinofuranosyl?1?4?[?-L-rhamnopyranosyl?1?2?]-?-D-glucopyranosyl-?-D-chacotriosyl-26-O-?-D-glucopyranoside?7?,3?-O-?-L-rhamnopyranosyl?1?2?-O-?-D-glucopyranosyl homo-aro-cholest-5-ene-26-O-?-D-glucopyranoside?8?,?25R??5?6?-ene-spirostanol-17?,3?-dihydroxy-3-O-?-D-glucopyranosyl-?1?4?-3-O-?-L-rhamnopyranosyl-?1?4?-[3-O-?-L-arabinofuranosyl-?1?2?]-?-D-glucopyranoside?9?,paris saponin VII?10?,?25R??5?6?-ene-spirostanol-17?,3?-dihydroxy-3-O-?-D-glucopyranosyl-?1?3?-[?-L-r hamnopyranosyl-?1?2?]-?-D-glucopyranoside?11?,paris saponin H?12?,paris saponin VI?13?,Parisyunnanoside F?1?.Among them,compound 6 is a new compound.Conclusion:PPC has significant anti-inflammatory analgesic action and polyphylla saponin compounds is the main composition of anti-inflammatory and analgesic action.Partial saponins have stronger hemolysis activity.The compounds from PPC have different degrees of cytotoxic activity and anti-inflammatory activity.And the above ground of PPC also showed analgesic anti-inflammatory activity,which worth to conduct in-depth research to realize the comprehensive utilization of PPC resources.
