Objective:To detect the expression of IDH1R132 H and cell proliferation antigen Ki-67 protein in non-tumor brain tissues and gliomas.To investigate the relationship between the expression of IDH1 and Ki-67 protein and the pathological grade of gliomas,and to identify IDH mutations in gliomas and brain glia.The relationship between the proliferation of tumor cells provides an important reference for glioma pathological grade and clinical individualized treatment.At the same time,it provides a part of the pathological grade of glioma IDH1R132 H mutation rate and Ki-67 expression in the glioma database in Jiangxi Province.Experimental data.Methods:Select paraffin-embedded specimens from 160 patients with cerebral gliomas from Department of Pathology,First Affiliated Hospital of Nanchang University from January 1st,2007 to September 30 th,2017,and 10 cases of normal cerebral tissue with intracranial decompression after cerebral contusion As a control group(frontal lobe or temporal lobe).Max VisionTM immunohistochemistry was used to detect the expression of IDH1R132 H specific antibody and Ki-67 protein in the IDH1 gene mutation.Combining the clinical data of glioma cases,the results were added to the analysis(see statistical methods below).Results :(1)The positive rate of IDH1 in glioma tissue was 40%(64/160).IDH1 expression was negative in the control group,and there was a significant difference in IDH1 expression between the control group and the glioma group(c2= 6.145,p<0.05).In addition to WHOI grade gliomas,the positive rate of IDH1 decreased with the increase of malignancy of tumor tissues.The positive rate of IDH1 in WHO II grade glioma tissues was 60%(41/68).WHO grade III gliomas The positive rate of IDH1 in tissues was 38%(20/53),and the positive rate of IDH1 in WHOIV grade glioma tissues was 12%(3/26).The difference in positive degree was statistically significant(c2 = 19.191,p<0.05).IDH1 has a high positive detection rate of 40%(64/160)in glioma tissue,but IDH1 expression is negative in the control group,and there is a difference in IDH1 expression between the control group and the glioma tissue.Significant(c2=6.145,p<0.05).Except for WHOI grade gliomas,the results showed that the positive rate of IDH1 decreased with the increase of malignancy of tumor tissues,and the positive rate of IDH1 in WHO II grade glioma tissues.At 60%(41/68),the positive rate of IDH1 in WHO grade III glioma tissue was 38%(20/53),and the positive rate of IDH1 in WHOIV grade glioma tissue was 12%(3/26)The difference between the positive levels was statistically significant(c2 = 19.191,p < 0.05).(2)The positive expression rate of Ki-67 in the control group was lower(LI<1%),and the positive expression rate in glioma tissue was significantly higher(P<0.05).WHOI level The Ki-67 labeling index(LI)of glioma tissues averaged 1.69%(95% CI: 0.63% to 2.75%),and the Ki-67 labeling index(LI)of WHO II grade brain glioma tissues averaged 7.38%.(95% CI: 6.71% to 8.05%).The average labeling index(LI)of Ki-67 in WHO III glioma tissue was 12.92%(95% CI: 11.73% to 14.11%).WHOIV grade brain glia The average labeling index(LI)of tumor tissue Ki-67 was 21.58%(95% CI: 17.56% to 25.59%),and the four groups had significant statistical differences(F=66.752 p<0.001).There is also a positive correlation between the expression of Ki-67 and the degree of malignancy of tumor tissues.The expression of Ki-67 in the IDH1R132 H mutant group was lower than that in the wild type(non-mutant)group(t=-11.116,P<0.001).The Ki-67 marker index of IDH1 R132 H mutant and wild-type glioma They were(6.77±3.206)and(17.66±7.152),respectively,suggesting that IDH1R132 H mutations reduce the proliferation activity of gliomas.(3)The Spearman correlation analysis was performed on the data.The analysis showed a negative correlation between the values of IDH1 and Ki-67(p<0.001,r=-0.562),and the results showed that the expression of IDH1 was correlated with the gender of patients with gliomas.No correlation(p>0.05)was associated with the patient's age,and IDH1 was more commonly expressed in glioma patients less than or equal to 45 years of age(p<0.05).Conclusion:(1)IDH1 and Ki-67 were positively expressed in different grades of glioma tissues.No expression of IDH1 and Ki-67 was found in the control group.IDH1 may be associated with gliomas.(2)The expression of IDH1 and Ki-67 is associated with the pathological grade of glioma?(3)Compared with the IDH1 negative expression group,the glioma proliferation activity was lower in the IDH1 positive expression group.
