Expression Of MiRNA-125b In Melanoma And Correlation With Prognosis

Objective:1.To detect the expression of miRNA-125 b in paraffin-embedded tissues of melanoma.2.To explore the relationship between the expression level of miRNA-125 b and the clinical features,diagnosis and prognosis of melanoma patients.Method:29 cases of melanoma tissue and 16 cases of nevus were collected from the first affiliated hospital of Nanchang University from January 2014 to August 2017.Clinical and pathological data are required to be complete.According to TNM staging standard(AJCC 2010),the specimens were divided into 3 groups: the first group of nevus(as Control),the second group: Phase I and Phase II,and third group: Phase III and Phase IV.Real-time fluorescence quantitative nucleic acid amplification assay(QRT-PCR)was used to detect the level of miRNA-125 b expression in each group of specimens.Using statistical software to analyze the correlation of miRNA-125 b expression in melanoma,such as age,sex,tumor size,lymph node metastasis,TNM staging and lesion site.ROC curve analysis miRNA-125 b the diagnostic value of melanoma.Kaplan-meier method and Log-rank test were used to analyze the survival rate of melanoma patients,and Cox analysis was performed to analyze whether miRNA-125 b expression was an independent predictor of poor prognosis of melanoma.Result:1.The expression level of miRNA-125 b in the primary group(0.781±0.442)and the transfer group(0.366±0.300)was lower than that in the control group(1.100±0.845),and the miRNA-125 b expression level of the transfer group was significantly lower than that in the primary group(p<0.05).2.The expression level of miRNA-125 b is closely related to lymph nodemetastasis(P= 0.001)and TNM staging(P=0.009),and has no significant correlation with sex,age,tumor size(diameter)and lesion site(p>0.05).3.The ROC curve shows that the area under the curve(AUC)was 0.786,and95% confidence interval was the 0.618-0.954(p<0.05).The survival period of miRNA-125 b low expression group was shorter than that in the High expression Group(p<0.05),and multivariate analysis showed that miRNA-125 b expression level may be an independent predictor of poor prognosis of melanoma.Conclusion:1.The expression of miRNA-125 b in patients with melanoma was decreased,and the abnormal expression was correlated with lymph node metastasis and TNM staging.2.The expression of miRNA-125 b is valuable for the diagnosis of melanoma.The expression of miRNA-125 b can be used as an independent predictor of prognosis in melanoma patients.
