Objectives:To observe the changes of GRP78 and TNF-?in acute lung injury induced by paraquat poisoning,to investigate the influence and significance of hydrogen sulfide on the expression of acute lung injury's endoplasmic reticulum stress protein GRP78of rats which is induced by paraquat..MethodsExperiment 1:50 S-D rats were randomly divided into 10 groups,including paraquat poisoning group?3h,6h,12h,24h,48h?,normal saline control group?3h,6h,12h,24h,48h?.Observation general view of lung tissue and pathological changes of lung tissue under light microscope,blood gas analysis of abdominal aorta,determination of protein content in bronchoalveolar lavage fluid by BCA,lung wet/dry weight ratio,determination of TNF-?in peripheral blood by ELISA,determination of endogenous H2S in serum by spectrophotometry,GRP78mRNA content in lung tissue was detected by fluorescence quantitative PCR,detection of GRP78 expression in lung tissue by Western Blot.20 S-D rats were randomly divided bleeding,ecchymosis,lung tissue fluid,bleeding,bleeding,blisters,poor elasticity,is most obvious in the48h group.1.2 HE staining under microscopeIn normal saline control group,the lung tissue structure was complete,the alveolar wall was complete,the alveolar space was clear,and there was no congestion,edema and exudation in the pulmonary interstitium.Inparaquat poisoning group,the damage of alveolar structure,alveolar inflammatory cells,red blood cell invasion;interstitial lung congestion,edema,exudation,pulmonary interstitial capillary dilatation,hyperemia,micro thrombosis,hyaline in lung tissue,decreased the level of serum TNF-?,and reduced the degree of lung injury.
