Objective:To study the relationship between AKR1B10 expression and the clinic characteristics of breast cancer and to explore the effect of AKR1B10 expression on the proliferation and migration of breast cancer cells and its mechanism.Methods:63 cases of breast cancer and para-carcinoma tissue specimens were collected from the First People's Hospital of ChenZhou from 2013 to 2016.The expression level of AKR1B10 in the tissue specimens was detected by immunohistochemistry.We analyzed the correlationship between AKR1B10 expression and clinical characteristics.In this study,AKR1B10 gene was cloned into MCF-7,MDA-MB-231 cells without AKR1B10 endogenous expression by lentivirus system and MCF-7/AKR1B10,MDA-MB-231/AKR1B10 cells with the stable expression of AKR1B10 were established.The expression of AKR1B10 was knocked down in MCF-7/AKR1B10,BT20 cells by AKR1B10 shRNA silence plasmid.The expression level of NF-?B P65 in the nuclear protein of MCF-7 cells and Phosphorylation level of NF-?B P65 in the total protein of MCF-7 and MDA-MB-231 cells were detected by western blot.The localization of NF-?B P65 in MDA-MB-231 cells was checked by cell immunofluorescence.The effects of AKR1B10 expression and NF-?B inhibitor on breast cancer cells were observed by cell clone formation experiment,MTT assay,scratch experiment and transwell experiment.Results:AKR1B10 was highly expressed in breast cancer and positively correlated with tumor size and lymph node metastasis.There was a positive correlation between AKR1B10 expresssion and tumor size(r = 0.324;p<0.05).Simultaneously,AKR1B10 expression was associated with lymph node metastasis(r = 0.324;p<0.05).AKR1B10 expression promoted the cell proliferation and migration of MCF-7 and MDA-MB-231 cells,up-regulated the expression leves of NF-?B P65 in the nuclear protein of MCF-7 cells and Phosphorylation level of NF-?B P65 in the total protein in MCF-7 and MDA-MB-231 cells.NF-?B inhibitor remarkably suppressed the cell proliferation and migration of breast cancer cells.Conclusion:1.AKR1B10 was highly expressed in breast cancer tissue and was positively correlated with tumor size and lymph node metastasis.2.AKR1B10 promotes the proliferation and migration of breast cancer cells by activating NF-?B signaling pathway.
