objective: Salinomycin Induced endoplasmic reticulum stress and investigate the possible mechanism of in prostate cancer cells for PC-3,to provide aexperimental basis for the clinical treatment of prostate cancer in Salinomycin.Materials and methods: Logarithmic growth phase PC-3 cells in prostate cancer were tested.(1)Detection of major gene expression in endoplasmic reticulum stress by q PCR:The experiment was divided into concentration gradient groups and time gradient groups.The concentration gradient group was 0,0.25,0.5,1,2,5 umol/mL in Salinomycin,dispose PC-3 cells in prostate cancer with 24h;The time gradient groups was 5 umol/mL in Salinomycin,dispose PC-3 cells in prostate cancer with0,4,6,8,12,24 h.(2)Detection of major gene expression in endoplasmic reticulum stress by Westren Blot: The experimental grouping method is the same as experiment(1).(3)Westren Blot was used to detect the expression levels of major proteins in endoplasmic reticulum stress after ATP2A3 overexpression and ATP2A3 interference.(4)Westren Blot was used to detect the expression levels of major proteins in endoplasmic reticulum stress after ATP2A3 overexpression and ATP2A3 interference that dispose PC-3 cells in prostate cancer with24 h in Salinomycin.(5)Using T test variance analysis of qPCR results,Using Single factor variance analysis of Westren Blot results.results :(1)Visible through qPCR results:0?0.25?0.5?1?2?5?mol/mL in Salinomycin dispose of prostate cancer PC-3 cells after 24 h,increased expression quantity of endoplasmic reticulum stress important genes BiP,PERK,eiF2 a,ATF4,IRE1 a,ATF6 all up in the role of Salinomycin,It is suggested that the endoplasmic reticulum stress in PC-3 cells of prostate cancer is dependent on the concentration of Salinomycin;5?mol/mL in Salinomycin dispose of prostate cancer PC-3 cells after0,4,6,8,12,24 h,increased expression quantity of endoplasmic reticulum stress important genes BiP,PERK,eiF2 a,ATF4,IRE1 a,ATF6 all up in the role of Salinomycin,It is suggested that the endoplasmic reticulum stress in PC-3 cells of prostate cancer is dependent on the time of Salinomycin.(2)Visible through Westren Blot results: 0?0.25?0.5?1?2?5?mol/mL in Salinomycin dispose of prostate cancer PC-3 cells after 24 h,increased expression quantity of ATP2A3 protein and endoplasmic reticulum stress important protein BiP,PERK,eiF2 a,ATF4 is dependent on the concentration of Salinomycin,IRE1 a,ATF6 not obvious in expression quantity change,the difference was not significant(p>0.05).5?mol/mL in Salinomycin dispose of prostate cancer PC-3 cells after 0,4,6,8,12,24 h,increased expression quantity of ATP2A3 protein and endoplasmic reticulum stress important protein BiP,PERK,eiF2 a,ATF4 is dependent on the time of Salinomycin,IRE1 a,ATF6 not obvious in expression quantity change,the difference was not significant(p>0.05).(4)Visible through Westren Blot results: The endoplasmic reticulum stress important protein PERK,eiF2 a,ATF4 not obvious in expression quantity change after ATP2A3 over expression group and ATP2A3 interference group,the difference was not significant(p>0.05).5?mol/mL in Salinomycin dispose of prostate cancer PC-3 cells after 24 h,increased expression quantity of endoplasmic reticulum stress important protein PERK,ei F2 a,ATF4 in ATP2A3 over expression group,the difference was significant(p<0.05);not obvious of endoplasmic reticulum stress important protein PERK,eiF2 a,ATF4 in ATP2A3 interference expression group,the difference was not significant(p<0.05).conclusion: Salinomycin may activate the Bip-PERK pathway by upregulating the expression of ATP2A3,inducing the endoplasmic reticulum stress of prostate cancer PC-3 cells.
