Curcumin Nicotinate Prevent Cognitive Impairment Regulatin Autophagy

[Backgroud and Objective] To determine curcumin nicotinateprevent diabetic cognitive dysfunction(DACD)in rats and investigates whether its protective effect involves regulation of hippocampal autophagy.[Methods]: The rats were randomly assigned to six groups:(1)Control group(n = 15),which was the normal rats and receivedphosphate buffer saline,(2)Vehicle group(DM)(n = 15),which was the diabetic rats and received phosphate buffer saline(3)DM + curcumin nicotinate groups(n =15),which were diabetic rats and treated with curcumin nicotinate(100mg/kg per d)for 28 days(4)DM + Rapamycin group(n=15),which were diabetic rats and treated with Rapamycin(1mg/kg)for 14 days,(5)DM+curcumin nicotinate + Chloroquine Diphosphate(n=15),which were diabetic rats treated with curcumin nicotinate(100 mg/kg per d)for 28 days and Chloroquine Diphosphate(40mg/kg)for 14 days(6)DM +curcumin nicotinate + 3MA(n=15),which were diabetic rats treated with curcumin nicotinate(100 mg/kg per d)for 28 days and 3MA(1.5mg/kg)for 14 days.Hematoxylin-eosin staining observe that the neurons significantly and the arranged disorder in hippocampus area.To determine whether curcumin nicotinate improves spatial learning and memory in diabetes mice,we have taken advantage of Morris water test to examined mice's cognitive performance.To determine the relationship between cognitive impairment in diabetes rats and autophagy,western blotting was carried out.LC3-?(microtubule-associated protein 1 light chain 3)is a reliable autophagy marker.p62,a protein associated with autophagosomes,which is degraded in the autolysosome,and is commonly regarded as autophagy substrateprotein.Bax is a protein associated with apoptosis.[Results]: 1)Curcumin nicotinate prevent cognitive impairment;2)Curcumin nicotinate prevent nerve cell from damaging in diabetic rats;3)Curcumin nicotinate prevent cognitive impairment regulating autophagy;4)Curcumin nicotinate reduce neurons apoptosis in diabetic rats.[Conclusion]: curcumin nicotinate may prevent DACD which is probably by regulating autophagy.
