Objective: To screen and anal yze the dif ferentially expressed circulatory molecules in m yocardial tissue in T OF patients by gene sequencing and bioinformatics methods,and to provide a basis for further study of the molecular mechanism and regulation of pathogenesis.Methods: 5 cases of right ventricular myocardium in children with tetralogy of Fallot?experimental group?,5 cases of right ventricular myocardium in children with ventri cular septal defect?control group?.Extraction of total RNA,After the sample is tested,Screening of differentially expressed circRN A using gene sequencing and bioinformatics.Analysis of gene s,The gene was enriched by KEGG Pathway,screening of pathways asso ciated with tetralogy of Fallot were screened.Results: Experimental group and control group contrast studies showed that the expression of circ RNA was up to 3238,the expression was down 17255,only 2191 were expr essed in experimental group.only 16375 were expressed in control group,and 4239 circ RNAs were co-expressed in the two groups are.Hg38circ008601,hg38circ0018611,Hsacirc0005087 significantly improved,hg38circ0002916,hg38circ0006601,hg38circ0019138,hg38circ0010081,hsacirc0002353 down significantly,The gene of circRNA was enriched and analyzed,and 32 signal pathways were screened out.Conclusion: 1.The gene expression profile of tetralogy of Fallot was obtained by gene sequenc ing and bi oinformatics methods,and t he differentially expressed circRNAs were screened and chromosomalized.2.The expression of circ RNA was up-regula ted by 3238,the expression was down-regulated by 17 255,and only 2191 were found i n the myocardium of patients with tetralogy of Fallot,and only 16375 were expressed in the myocardium of ventricular septal defect.There are 4239 circRNAs.3.The enrichment analysis of the source gene,com bined with the relevant literature found that adhe sion spots,phosphoinositol pathway may be associated with outflow tract l esions,TGF-? signaling pathway may be related to the pathogenesis of TOF.
