Objectives:To explore the effect of HMGB1 on cell cycle,autophagy and chemosensitivity of bladder cancer cells and the mechanism by flow cytometry,Western Blot and CCK-8 detection means to detect bladder cancer cell line(BIU-87 and T24)HMGB1 loss of function model(p Talen-HMGB1 function model,HMGB1 si RNA functional model)datas.Methods:The first part: endogenous detection:1.The expression of HMGB1 in different TNM staging and different grading tumors was detected by using bladder cancer microbar-ray.2.The mRNA levels of HMGB1 were measured by using bladder cancer cell lines BIU-87 and T24 cells.The second part: function modeling1.HMGB1 functional loss model(p Talen-HMGB1 functional model,HMGB1 si RNA functional model).2.To construct and construct HMGB1 target knockout plasmid pairs,transfected bladder cancer cell lines BIU-87 and T24,respectively,to screen stable cell lines.3.To measure cellular models HMGB1 expression levels by WB.The third part: functional experiment:1.Cell cycle was measured using flow cytometry.Different groups: the NC group,The Con group,the scrambled si RNA group,the HMGB1 si RNA group,the p Talen NC group,the p Talen-HMGB1 group.2.Detect the changes of LC3 B II / I and Beclin 1 protein expres-sion levels to reflect the changes of autophagy using WB.Different groups: the NC group,The Con group,the scrambled si RNA group,the HMGB1 siRNA group,the p Talen NC group,the p Talen-HMGB1 group.3.Detected the effect of cisplatin on the growth of two cells using CCK-8.Different groups: the NC group,The Con group,the scrambled siRNA group,the HMGB1 siRNA group,the p Talen NC group,the pTalen-HMGB1 group.Results: 1.The cell cycle of bladder cancer cells T24 and BIU-87 was inhibited by flow cytometry.The cell cycle of bladder cancer cells T24 and BIU-87 after transfection of HMGB1 siRNA was also inhibited.2.The levels of autophagy in BIU-87 and T24 cells were decreased after transfection of HMG1 cells by transfection of p Talen-HMGB1 plasmid.3.The levels of autophagy in BIU-87 and T24 cells were also decreased.CCK-8 was transfected into pTalen-HMGB1,and the sensitivity of BIV-87 and T24 was increased,and the sensitivity of transfected HMGB1 si RNA was higher than that of HMGB1 siRNA group.Conclusions:1.Cell cycle was inhibited of bladder cancer cell lines BIU-87 and T24 after HMGB1 knockout/silencing.2.Autophag-y was decreased of bladder cancer cell lines BIU-87 and T24 after HMGB1 knockout/silencing.3.Chemosensitivity was increased of bladder cancer cell lines BIU-87 and T24 after HMGB1 knockout/silencing.
