Poor living habits and stress lead to an increase in the incidence of acute myocardial infarction,and become one of the main killers of health.The current method of treating the disease is preferred by percutaneous coronary intervention(PCI)to recanalization the occluded vessels.Despite the epicardial coronary blood flow was successfully restored of myocardial infarction patients,a significant ratio of patients with myocardial infarction is still suffering from severe microvascular perfusion injury.This reperfusion failure at the microvascular level is a condition known as no-reflow(NR)or microvascular injury.NR is one of the independent clinical predictors of AMI,and is associated with an increased risk of heart.The underlying pathophysiology of microvascular injury is complex and multifactorial and is not fully understood.Limb ischemia is simple and easily being accepted has been widely studied and is considered to be one of the most effective treatments for ischemic heart disease.Our group has confirmed that limb PerC could reduce myocardial NR phenomenon in rats.And also verified that the metabolites of Lactic acid(LA),Uric acid(UA),1,6Fructose Diphosphate(FDP)maybe the potential biomarkers of myocardial NR phenomenon and limb PerC.Objective This study aim to study the relationship between no-reflow area and dynamic changes of serum Lactate acid,Uric acid,Urea nitrogen levels and 1,6 Fructose Diphosphate.To provide new ideas and references for the pathogenesis and treatment of reflow.Methods In this study,the rat model of myocardial ischemia reperfusion injury was prepared by ligating the left anterior descending coronary artery(LAD).Sixty healthy male SD rats were used for this study,all rats divided into 7 groups by accident,namely 15 min myocardial ischemia group,30 min myocardial ischemia group,45 min myocardial ischemia group,60 min myocardial ischemia group,90 min myocardial ischemia group,sham operation group and PerC group.All the groups were fixed in 2 hour reperfusion.The sham operation group rats did not undergo occlusion of LAD.The PerC group rats were ischemic 45 min and reperfusion 2h,and before reperfusion,the rats leg underwent five ischemic conditioning cycles of 30 s ischemic and 30 s reperfusion.No-reflow area of myocardium,infarction area and the area of dangerous were obtained by staining with thioflavin S,Evans blue and TTC respectively.The changes of histopathological features were observed by HE staining in the heart and kidney tissues.The dynamic changes of serum LA,UA,BUN and FDP were measured by arterial blood sampling.Results(1)Except for 15 min myocardial ischemia group,the rest of the ischemia reperfusion groups showed significant myocardial NR phenomenon.The area of NR area and myocardial infarct size enlarged with the expansion of ischemia time,and the boundary between the NR area and the blood perfusion area was distinct,and the infarct area,non-infarct area were clear and the risk area was approximately the same.(2)There was some positive correlation between no reflow area of myocardial ischemia reperfusion injury and the levels of serum LH,UA,BUN,FDP in rats.(3)When the time of ischemia was 45 min and the time of reperfusion was 2h,the PerC group area of no-reflow and infarct area and the levels of serum LH,UA,BUN were lower than those non-PerC group,but there was no significant statistical difference in serum level of FDP.Conclusion The area of no-reflow enlarged with the expansion of ischemia time,and there was some positive correlation between no reflow area and the levels of serum LH,UA,BUN,FDP in rats...
