Background:Chronic heart failure is the final destination of most cardiovascular diseases,which is a serious threat to human health.It has high incidence,high disability rate and high mortality.In recent years,the incidence of the disease is gradually increasing,which has become a serious public health problem.To correct cognize it take a correct treatment and preventive measures have become an urgent problem.The researchs have proved that ventricular remodeling is the main pathological mechanism of CHF,and myocardial fibrosis plays an important role in the development of ventricular remodeling.To inhibition the myocardial fibrosis can improve the development of ventricular remodeling effectively,which can slow down the progression of chronic heart failure.Preliminary study had prove that optimal preseription of Jiashen can restrain the ventricular remodeling and myocardial fibrosis.Optimal preseription of Jiashen could treat CHF better.But its mechanism remains to be studied.Objective:we observed the Influence of optimal preseription of Jiashen on heart function,Brain natriuretic peptide,angiotention?,hydroxyproline and distribution of collagen in myocardial of rats which heart failure after myocardial infarction.discuss the Influence of the optimal preseription of Jiashen on can restrain the ventricular remodeling and myocardial fibrosis.And to discuss whether the above function of the prescription of Jiashen is related to the inhibition of the expression of Ang II.Method:Seventy healthy adult rats were randomly divided into the operated group?n=60?and the sham-operated group?n=10?.The operated group was induced by left anterior descending coronary artery liation;The sham-operated group underwent the same method,but the suture was not tied.The rats surviwed to the 24 hours after MI were feed for 4 weeks.Do color Doppler ultrasound,32 rats of the EF<50%were randomly divided into 4 groups?n=8 in each group?:The model group of MI?model group for short?,Low dose group of optimal preseription of Jiashen?low dose group for short?,high dose group of optimal preseription of Jiashen?high dose group for short?,the losartan group.chose sham operation group?n=8?.The sham-operated group and the model group were administrated pure water(1ml·100g-1·d-1),the low dose group was given the optimal prescription of Jiashen(3g·kg-1·d-1),the high dose group was given the optimal prescription of Jiashen(6g·kg-1·d-1),the losartan group was given the losartan(30mg·kg-1·d-1).Intragastric administration was given once a day for 4 weeks,Then we observed general condition of all of rats such as diet,activity,pelage and so on.4 weeks later,do the color doppler ultrasound to detect the left ventricular end systolic diameter?LVESD?,the left ventricular end diastolic?LVEDD?,the interventricular septum thickness?IVS?,the left ventricular fractional shortening?LVFS?and the left ventricular ejection fraction?LVEF?.To collect the abdominal aorta blood for testing serum BNP by enzyme-linked immunosorbent assay?ELISA?,execute the rats after weighing?BW?,measure the left ventricular mass index?LVMI?,collected the myocardial tissues,to detect the HYP content in myocardial tissue quantitative collagen content by the method of alkaline water,to observe the distribution of collagen in myocardial tissue by the method of masson,to detect the Ang II content in myocardial tissue by ELISA.Results:The research showed that:?1?The rats in sham operation group had better mental,activity,diet and weight gain.Other groups of rats had varying degrees of dizziness,fatigue,poor vigilance,bending fatigue,dull hair or hair loss,the weight increased slowly,do not think of fresh water,loose stools,diarrhea and so on.?2?The results of color doppler ultrasound:Compared with the sham operation group,the weight increased slowly,the rats do not think of fresh water,loose stools,diarrhea anon group,the LVESD and LVEDD were significantly increased in model group and each drug group,the IVS,LVEF and LVFS were significantly lower?P<0.05,P<0.01?.Compared with the model group,the LVEDD in high-dose group and losartan group,the LVESD in each drug group were significantly lower?P<0.05,P<0.01?,the IVS in each drug group,the LVEF and LVFS in high-dose group and losartan group were significantly increased?P<0.05,Pred w<0.01?.?3?The results of serum BNP:Compared with sham operation group,the model group and each drug group were significantly higher?P<0.05,P<0.01?.Compared with the model group,each drug group was significantly lower?P<0.05,P<0.01?.?4?The results of the LVMI:Compared with the sham operation group,the model group was significantly higher?P<0.05?.Compared with the model group,the high-dose group and losartan group were significantly lower?P<0.05?.?5?The results of the HYP in myocardium:Compared with sham operation group,the content of HYP in myocardium of model group and each drug group was significantly higher?P<0.01?.Compared with the model group,the high-dose group and the losartan group were significantly lower?P<0.05?,and the effect of the prescription of Jiashen was dose-dependent.?6?The results of the distribution of collagen in myocardium:In the sham operation group,the normal myocardial cells were bright red color,arranged orderly.Collagen fibers are distributed in myocardial cells.The collagen fibers in model group was significantly higher than that in sham operation group.They are woven into a net,piled up into pieces.Compared with the model group,the distribution of collagen in the each drug group was significantly reduced,and the effect of high-dose and losartan was similar.?7?The results of the Ang II in myocardium:Compared with sham operation group,the Ang II was significantly higher in model group?P<0.01?.Compared with the model group,the high-dose group and losartan group were significantly lower?P<0.01?.Conclusion:?1?The rats with heart failure after myocardial infarction,the LVEDD and LVESD increased significantly,the IVS decreased significantly,the LVEF and LVFS significantly decreased,the serum BNP,the content of HYP and the content of Ang II in myocardium ware significantly increased,It is suggested that the heart function of rats with heart failure after myocardial infarction became lowering,it occurred the ventricular remodeling and myocardial fibrosis.?2?The optimal preseription of Jiashen can reduce can reduce the LVEDD,LVESD,elevated the IVS,LVFS and LVEF,reduce the serum BNP,the LVWI,the content and distribution of collagen and the content of Ang II in myocardium in rats with heart failure after myocardial infarction,play the roles of improving the heart function,inhibitting ventricular remodeling and reducing the myocardial fibrosis.The above functions were related to the inhibition of Ang II.?3?The influences of the optimal preseription of Jiashen on cardiac function,ventricular remodeling and myocardial fibrosis are the same to losartan.
