| Objective: To survey related signaling pathways of the changes of key proteins Notch-1,Wnt3 a,ERK1/2 and the role of tonifying kidney recipe in the natural aging rats,and to explore the mechanism of aging and tonifying kidney recipe.Method: The models of our experiment were SD male rats aged 24 months old.30 male rats were divided into three groups randomly(10 rats per group),elderly group,Zuogui Pill group,Yougui Pill group;the three groups of rats were all 24 months old.Another 10 were only 5 months for young group.Zuogui Pill group and Yougui Pill group from the beginning of the 20 months,the dosage of gavage was the Zuogui Pill and Yougui Pill 0.675g/100g/d(equivalent to 15 times of adult clinical dosage),stopped two days a week,lasting for four consecutive months.All the groups of rats were supported with normal diet and drinking water.The expression of key proteins,Notch-1,Wnt3 a and ERK1/2 in hippocampus of the natural aging rats were detected by Western blot method;Immunohistochemical method was used to detect the expression of key proteins,Notch-1 and ERK1/2 in dentate gyrus of hippocampus of the natural aging rats.Results: 1.Compared with the young group,the expression of Notch-1,Wnt3 a and ERK1/2in the hippocampus of the elderly group was decreased(P<0.05);Compared with elderly group,the expression of Notch-1 and Wnt3 a in the hippocampus of the the Zuogui Pill group and Yougui Pill group was increased(P>0.05).Compared with elderly group,the expression of protein ERK1/2 of Zuogui Pill group was increased,but the expression of protein ERK1/2of Yougui Pill group was decreased.2.Compared with the young group,the expression of Notch-1 and ERK1/2 in the dentate gyrus of hippocampus of elderly group was decreased(P<0.05);compared with elderly group,Zuogui Pill group and Yougui Pill group could promote the protein expression of Notch-1 and ERK1/2 in the dentate gyrus of hippocampus(P<0.05).Conclusion: 1.The expression of protein Notch-1,Wnt3 a and ERK1/2 in aged rats hippocampus was decresed,Zuogui Pill and Yougui Pill could increase the expression of protein Notch-1 and ERK1/2;2.The possible mechanism of Zuogui Pill and Yougui Pill can delay the senility may be that they can regulate the signaling pathways of Notch and MAPK. |
