Objective: To investigate the expression of CD4 +,CD8 + T cells and tertiary lymphoid structure in breast cancer tissue and its clinicalMethod:(1)A total of 150 cases of paraffin-embedded tissue sections of invasive ductal carcinoma(73 cases of pre-study specimens and 77 cases of late supplemental samples)were collected from May 2010 to January 2016,(2)Experiment Part I:1)Group: TLS negative expression groupwas set as control group(group A),TLS positive expression groupwere experimental group(group B).2)The tissue samples were successively sliced and numbered 1 to 11 in sequence;3)All tissue sections numbered 1 were incubated with PBS and incubated with negative antibody for negative observation.All tissue sections numbered 2 to 9 were added with CD3,CD4,CD8,HMGB1,CD62 L,CD20,CD21 and BCL-6respectively.Then,They were incubated with secondary antibodies for visual observation.The expression rates of CD3 + T cells,CD8 + T cells,CD4 + T cells,HMGB1 protein and TLS were measured.Number 10,11 spare.(3)The second part of the experiment:1)Group: the control group for the A 'group: characterized by TLS negative,and CD3,CD4,CD8 were negative.The experimental group was B ',C': B 'group: characterized by TLS positive,group C': characterized by TLS negative,CD3 positive,CD4 and / or CD8 positive.2)Comparison of the differences in the age,tumor diameter,lymph node metastasis,pathological grade,and pTNM staging of the A ',B' and C 'groups.(4)All the data in this study were analyzed by SPSS18.0 statistical software: the difference analysis was analyzed by chi-square test,and the correlation analysis was analyzed using pearson correlation analysis.By ?=0.05 for the level of test,P <0.05 is considered statistically significant.Result:(1)The positive expression rate of TLS in breast cancer was 41.3%(62/150),the negative expression rate was 58.7%(88/150).(2)The positive expression rate of CD8 in group A was 28.4 %(25/88)and 71%(44/62)in group B.Compared with group A,the positive expression rate of CD8 in group B was significantly higher than that in group A(P = 0.000 < 0.05).(3)The positive expression rate of CD4 in group A was 15.9 %(14/88),and the positive rate of CD4 in group B was 62.9%(39/62).Compared with group A,the positive rate of CD4 expression in group B was significantly higher than that in group A(P = 0.000 <0.05).(4)The high expression rate of HMGB1 in group A was 91%(80/88)in group A and 37.1%(23/62)in group B.Compared with group A,the positive expression rate of HMGB1 protein in group B was significantly lower than that in group A(P =0.000 <0.05).(5)The high-medium ratio of A ',B 'and C' was 32% in group A ',92% in group B',60% in group C '(P<0.05).Compared with the A 'group,the B' and C 'groups were significantly increased,and the difference was significant(P<0.05).Compared with the C 'group,the B' group was significantly higher than the B 'group(P <0.05).(6)The no lymphatic metastasis ratio of A ',B 'and C' was 7% in group A ',77% in group B',47% in group C '(P <0.05).Compared with the A 'group,the B' and C 'groups were significantly increased,and the difference was significant(P<0.05).Compared with the C 'group,the B' group was significantly higher than the B 'group(P <0.05).(7)The early and middle term(stage I + II)ratio of A ',B',C 'was 24% in group A',95% in group B 'and 53% in group C'(P <0.05),and there was significant difference between the B 'group and the C' group(P<0.05),and the difference was significant(P <0.05).(8)There was no significant difference in the composition of the three groups(P> 0.05)between the A ',B' and C 'groups on the age of the patients and the size of the breast tumor.Conclusion:1.High expression of CD4,CD8 andlowexpression of HMGB1 may be related to the good prognosis of patients with TLS positive.2.Comparedwith TLS-negative patients,TLS-positive breast cancer patients may have a betterprognosis.3.In TLS-negative breast cancer patients,CD4,CD8-positive expression may suggest a better prognosis.
