Part 1:The Study Of Cell Therapy For Solid TumorsObjective:A549 lung adenocarcinoma tumor bearing mice were used as experimental materials to investigate the effects and mechanisms of DC-CIK,hUCMSCs,and LR-MSCs on lung adenocarcinoma solid tumor alone or in combination with three kinds of cells.Methods:A total of 25 tumor bearing mice with A549 lung adenocarcinoma were established and all of them were randomLy divided into 5 groups,each group consisted of 5 nude mice:hUCMSCs group,DC-CIK+hUCMSCs group,hUCMSCs+DC-CIK group,LR-MSCs group and negative control group,and the corresponding cells were injected by tail vein administration.h UCMSCs was 1×10~5/0.1mL/only,DC-CIK was 1×10~6/0.1mL/only,LR-MSCs was 1×10~5/0.1mL/only.At the end of one week after treatment,the length of the transplanted tumor was measured,and the tumor volume was calculated.At the same time,the tumor-bearing mice were sacrificed and the tumor inhibition rate was calculated after intactly stripped tumor-bearing.The tumor tissues were examined by HE staining,and the expressions of VEGF,Bcl-2 and MMP-9 were detected by immunohistochemistry.Results:1.According to the volume and free dissociation quality of the solid tumor,the control group of each treatment group decreased.2.Each treatment group had significantly reduced the role of VEGF,bcl-2protein expression in the tumor tissue(P<0.05).There was no statistically significant difference between the treatment group hUCMSCs and the LR-MSCs treatment group.DC-CIK+hUCMSCs sequential combination therapy has the ability to lower the expression of VEGF,bcl-2 protein,in contrast to the combination of hUCMSCs+DC-CIK sequential therapy.The sequential combination therapy of DC-CIK+hUCMSCs is more powerful than the individual hUCMSCs therapy,which is a reduction in the expression of VEGF and bcl-2.The immunohistochemical results of MMP-9 in transplanted tumor were different,but there was no significant difference between these groups.Conclusion:1.The isolation of cell therapy and combination therapy have inhibitory effect on the tumor of lung cancer,and for the first time,LR-MSCs has been found to have an anti-tumor effect on lung adenocarcinoma.2.The cell combination therapy of inputting into DC-CIK before hUCMSCs had stronger antitumor effect than cell-combination therapy of inputting into hUCMSCs before DC-CIK.3.The sequential combination therapy of DC-CIK+h UCMSCs has the ability to inhibit the expression of VEGF,bcl-2 protein in solid tumor tissue than single h UCMSCs.Part 2 Clinical Practice Analysis of Ovarian Cancer Cell TherapyObjective:To discuss the efficacy and safety of cell therapy in patients with ovarian cancer.Methods:A postoperative volunteer.After signing informed consent,the general examination,the Karnofsky score,and the ECOG scores were given before treatment.Follow these procedures to enter the treatment:On the first day,the patient's blood was taken and the CIK cells were prepared by standard operatin.When the cells were cultured for two weeks,they were returned to the patient by intravenous fluids.A week later,the h UCMSCs were prepared by standard operating procedure and entered the patient hrough intravenous fluids.The above procedure is a course of treatment and with two courses of treatment.Results:After treatment,the patient's score were not significantly worse: 80-90;ECOG scores improved before treatment,from 1-2 to 0-1;Kidney function and blood sugar damage caused by chemotherapy prior to treatment are also normal after treatment.he ALP was reduced before treatment,Before and after treatment respectively:189.16 U/L,177.78U/L.CA12-5,HE-4,had a certain growth before treatment,before treatment respectively: 149.6 U/m L,196.4 pmol/L,and after treatment respectively: 296.3 U/m L,467.7 pmol/L.Conclusion : Cell therapy in the treatment of patients with nest cancer,can improve the physical condition of patients,and repair the adverse reactions caused by chemotherapy,enhance the immunity of patients with cancer progression schedule,but the long-term survival rate and should be further studied.
