Objective: To investigate the mechanism of CHK2 in the pathogenesis of lung squamous cell carcinoma and its drug resistance.To compare the expression of CHK2 in lung squamous cell carcinoma and adjacent tissues,and to analyze the relationship between CHK2 and age,lymph node metastasis,smoking,And to analyze the relationship between CHK2 and prognosis of lung squamous cell carcinoma,to observe the changes of intracellular CHK2 expression after lung squamous cell carcinoma H226 cell line was administered by cisplatin.Methods: 58 cases of lung squamous cell carcinoma were collected from October 2013 to March 2015 in Hunan Provincial Tumor Hospital and 17 cases of adjacent tissues were collected from the 58 cases.All of them were identified by the pathology department of our hospital,58 specimens were derived from surgical specimens,and all lung cancer patients were not received any chemotherapy or radiotherapy for the tumor,we use the method of immunohistochemistry(SP)detect the expression of CHK2 in the above tissues,and analyze relationship between the expression of CHK2 and the clinicopathological parameters of lung squamous cell carcinoma.Cell culturewas performed with lung squamous cell carcinoma H226 cell line,the effect of cisplatin on the proliferation of lung squamous cell carcinoma H226 cell line was detected by MTT,the expression of CHK2 protein was detected by immunocytochemistry.The above data were analyzed by spss19.0.Result: 1.CHK2 protein is mainly expressed in the nucleus and cytoplasm of lung squamous cell carcinoma.The positive rate of CHK2 protein in the cancer tissue was 11 cases,47 cases were negative,the positive rate was 19.0%(11/58),12 cases were positive in CHK2 protein,5 cases were negative,The positive expression rate was 70.5%(12/17),the difference was statistically significant(p<0.001);2.There was no significant correlation between CHK2 expression level and age,sex,lymph node metastasis,smoking,clinical stage and pathologic differentiation of lung squamous cell carcinoma(p > 0.05);3.There was no significant correlation between CHK2 expression and no progression-free survival(PFS)and total survival(OS)in patients with lung squamous cell carcinoma(p>0.05);The expression of CHK2 had no significant correlation with PFS and OS in stage ? and ?patients with lung squamous cell carcinoma(p>0.05),There was no significant correlation between the expression of CHK2 and PFS and OS in patients with stage ? squamous cell carcinoma(p>0.05);4.Cisplatin can inhibit the proliferation of H226 cells in a concentration-dependent manner;The cells were treated with different concentrations(0.5,1,2,4,8mg/l)cisplatin(corresponding to A,B,C,D,E groups)for 48 h,and theintracellular CHK2 protein The expression of CHK2 protein in the experimental group was higher than that in the negative control group,and there was no significant difference between the negative control group with the A and B groups(p > 0.05),and there was significant difference with the C,D and E groups(p <0.05).Conclusion: 1.CHK2 expression in lung squamous cell carcinoma was significantly down-regulated.2.Cisplatin could induce the up-regulation of CHK2 protein in lung squamous cell carcinoma H226 cell line.
