Objective:To investigate the relations of Smad7 and Smad7 gene rs4464148 SNP with cholangiocarcinoma cancer risk and prognosis.Methods: 70 patients' bile duct carcinoma tissue(bile duct carcinoma group)and 40 cases of bile duct tissues in patients without tumor diseases(control group)specimens were taken from Hunan Province People's Hospital between January 2013 and January 2014,all patients with bile duct cancer who had not received preoperative adjuvant chemoradiotherapy.checking the Smad7 expression in cholangiocarcinoma group and the control group with immunohistochemical staining SP method,further analysis of relationship of Smad7 positive expression rate with bile duct cancer clinical pathological were made;Smad7 gene rs4464148 polymorphism of 70 cases of bile duct carcinoma and 40 cases of bile duct tissue were detected with LDR-PCR(ligase detection reaction-polymerase chain reaction),then we analyzed relationship between different genotypes and bile duct cancer risk and its prognosis.Results:(1)The Smad7 positive expression rate of bile duct carcinoma group was obviously higher than that of control group(P < 0.05).(2)Rs4464148 Smad7 gene loci included AA,AG,GG three genotypes,and AG/GG genotype frequency and G allele frequency of bile duct carcinoma group were higher than the control group,which differences were statistically significant(P < 0.05).(3)The Logistic multifactor regression analysis indicated that AG and AG/GG genotype OR value of 1.412?1.354 respectively,with P values were 0.023,0.041,which indicated AG/GG genotype may be a bile duct cancer factors(4)AG/GG genotype was closely related to the Smad7 positive expression,and genotype frequency binded to lymph node metastasis?TNM ?/ ?period,have a relapse or progress group was obviously higher than that without lymph node metastasis,TNM ?/ ?period,no recurrence or development group,and the difference had a statistical significance(P < 0.05).(5)The survival analysis showed that 3 years survival rate in patients with rs4464148 AG/GG genotype than in patients with AA genotype,but there was no significant difference(P > 0.05).Conclusion:(1)The Smad7 positive expression rate was significantly higher in the bile duct carcinoma tissue contrast to bile duct tissues.(2)The Smad7 gene rs4464148 AG/GG genotype which closely related to the Smad7 positive expression,may be a bile duct cancer risk factors,bingding bile duct cancer lymph node metastasis and recurrence or progress and the later clinical TNM stages.It also had the potential to be one of the important indices for bile duct cancer prognosis.
