Background:Premature ovarian failure(POF)refers to the ovarian function failure which leads to amenorrhea of women before the age of 40.The incidence of POF(premature ovarian failure)is about 0.9?1.2%in women.Autoimmune disease,genetic disease,virus infection and the chemotherapy of malignant tumor all can lead to the insufficiency of basal follicle pool reserve,follicular artesian acceleration and ovarian function decline.The studies have shown that chemotherapy drugs can reduce the number of primary follicles by direct and indirect mechanisms until premature ovarian failure occurs.In clinical,the most common chemotherapy drugs that cause premature ovarian failure are alkylation,platinum compounds,anthracoid antibiotics,and alkanes.So using stem cell to treat the ovarian premature failure has huge clinical significance.Objective:To establish animal models of POF in mice with two different chemotherapy drugs.Explore the effect of UCMSCs treatment in ovarian failure caused by the different chemotherapy drugs.Methods:Cyclophosphamide(CTX)(80 mg/kg/d*8 d)and Cisplatin(CDDP),(3 mg/kg/d*3 + 2 mg/kg/d*5 d)to establish two different ICR mice models of premature ovarian failure.Comparing the weight,ovary morphology and number of follicles of different chemotherapy groups in mice,we estimate the damage degree of ovaries caused by different drugs.After 14 days,we inject human umbilical cord mesenchymal stem cells through tail intravenous.And two weeks later,we record the change of the body weight in mice,observe the ovarian morphology.Four mice are sacrificed for the right ovaries.Using routine pathological method,we record the change of the number of ovarian follicles by optical microscope.Results:The results showed that both CTX and CDDP mice had significantly lower body weight(P<0.05),and the size of ovaries in the chemotherapy groups are significantly shrinking compared to the blank groups.Counting the number of follicles in chemotherapy groups,we find different types of follicles decreasing and CDDP group showed a greater toxicity.After the umbilical cord mesenchymal stem cell treatment,the weight and the number of visible follicles increased,especially the primordial follicles(P<0.05).So we prove the UCMSCs can partially improve the ovarian damage caused by two chemotherapy drugs.Conclusion:1.Our study established the successful animal model of premature ovarian failure induced by the CTX and CDDP.Both chemotherapy drugs can significantly affect mouse ovaries and the development of follicles;2.The UCMSCs treatment can significantly reduce the damage on ovarian caused by two chemotherapy drugs.
