ObjectiveTo investigate the expression of HLA-DQB1-AS1 in cervical cancer and non-tumour tissues,and to investigate the corelation between HLA-DQB1-AS1 and the development of cervical cancer.MethodsThe expression of HLA-DQB1-AS1 in cervical cancer and non-tumour tissues was detected by TaqMan probe real-time quantitative polymerase chain reaction.A series of biological experiments,including bioassay analysis,luciferase reporter assay,CCK-8,platelet colony formation,transwell and nude mice tumor formation experiments,were performed for functional verification.ResultsTaqMan probe real-time quantitative polymerase chain reaction technology found that the expression level of HLA-DQB1-AS1 in cancer tissues was significantly higher than that in non-tumour tissues,and the difference was statistically significant(P <0.05).Biological analysis and luciferase reporter assay showed that HLA-DQB1-AS1 can competes with HLA-DQB1 mRNA in binding to miRNA-205-5p,and improve the expression of HLA-DQB1.In addition,CCK-8,platelet colony formation,transwell and nude mice tumor formation experiments showed that overexpression of HLA-DQB1-AS1 could promote cell proliferation,migration,invasion and tumor growth.ConclusionsHLA-DQB1-AS1 as a competing endogenous RNA,can promote the development of cervical cancer via competitive combination with miRNA-205-5p.
