Objective: To study the expressione of Beclin-1,LC3 and AQP4 protein in cortex and hippocampus of rats with cerebral ischemia-reperfusion injury and the efficacy of Edaravone and to further explore whether cerebral ischemia-reperfusion injury could induce autophagy occurrence,and the effection of intervention group on autophagy related protein and AQP4 protein.Methods: Sprague Dawley rats(n=32)were randomly divided into sham group,ischemia-reperfusion modle group and Edaravone group.Model group was subjected to middle cerebral artery occlusion(MCAO)of 2 h followed by reperfusion of 24 h.Animals assigned to sham group were separated common carotid artery,except that the thread was not advanced to the middle cerebral artery.Edaravone was injected intraperitoneally at a dose of 10 mg/kg at 15 min before reperfusion.The condition of nerve injury was conducted by Neurobehavioral score.The extent of brain infarction focus and the success of model were determined based on 2,3,5-triphenyltetrazolium chloride(TTC)staining.Neuropathology changes of neuron in cortex and hippocampus were observed using hematoxylin and eosin(HE)staining.The ultrastructure of brain tissue was observed by electron microscope.Moreover,The expression of Beclin-1,LC3 and AQP4 in cortex and hippocampus were measured by immunohistochemistry and Western Blot.Results: Neurobehavioral score shown that Neurological damage symptoms were obviously appeared in the model group after 24 h of cerebral ischemia-reperfusion(P<0.05),while the symptoms was alleviated in the intervention group(P<0.05).TTC staining results showed that the infarction focus was obviously observed in the model group after 24 h of cerebral ischemia-reperfusion,infarction focus was reduced in the intervention group.HE staining shown that the disordered arrangement and nucleus pycnosis in cortex and hippocampus neurons were observed in the model group after 24 h of cerebral ischemia-reperfusion,compared with sham group,and the injury of neuron was reduced in the intervention group.Immunohistochemical results shown that compared with the sham group,the expression of Beclin-1,LC3 and AQP4 protein were increased in the model group after 24 h of cerebral ischemia-reperfusion(P<0.05),and the expression of Beclin-1,LC3 and AQP4 protein were decreased in the intervention group(P<0.05).Western blot results were consistent with immunohistochemical results.Conclusion: The neurons damage were observed after cerebral ischemia-reperfusion.The increased expression level of Beclin-1,LC3 and AQP4 were observed after cerebral ischemia-reperfusion injury,which indicated that autophagy might be induced after cerebral ischemia-reperfusion injury and the relation of AQP4 and autophagy may be exist.Edaravone might reduce brain damage through regulating expression of autophagy-related protein and AQP4.
