Objective1.To explore the association between the five SNPs of angiotensin II receptor type 1gene with simple obesity and central obesity.2.To discuss the potential relationship among gene-gene interaction of the five SNPs and simple obesity and central obesity.3.To detect the relevance between AGTR1 haplotypes and simple obesity and central obesity.4.To discuss the possible interactive effect of AGTR1 haplotypes and environmental factors on simple obesity and central obesity.Methods1.Participants were recruited under the framework of the Prevention of Multiple metabolic disorders and Metabolic syndrome cohort study in Suzhou Industrial Park.A total number of 1591 subjects were randomly selected and genotyped for the five polymorphisms.Questionnaire survey and biochemical detection were also carry out.2.The association between AGTR1 SNPs and simple obesity and central obesity were analyzed by linear regression model.3.MB-MDR was used to explore the possible gene-gene interactions among AGTR1 SNPs.4.Haplo.score was used for the association between the two AGTR1 haplotypes and simple obesity and central obesity.5.Haplo.glm was used for the interaction between AGTR1 haplotypes and environmental factors on simple obesity and central obesity.Result1.People who carry rs2638360 mutant allele?TC+CC?have higher BMI and WC compared with wild type?LL?,mean difference and 95%Confidence interval was0.54?0.190.88?kg/m2 and 1.11?0.162.06?cm.Carriers in mutation of rs2638360seem to increased simple obesity and central obesity risk,OR and 95%Confidence interval was 1.44?1.101.90?and 1.36?1.021.83?.The other four SNPs of AGTR1gene did not show any significant association with BMI,WC,simple obesity and central obesity.2.There are significant gene-gene interactions among the SNPs of AGTR1 gene?P<0.05?.We found that 2 to 7-locus models have positive correlation with simple obesity,3-locus model have positive correlation with central obesity and 2,4,5-locus model have negative correlation with central obesity.3.Among the haplotypes consist of rs2638360 and rs2933249,C-T haplotype was positively associated with simple obesity?Score=2.39,P=0.011?and was not significantly associated with central obesity.Among the haplotypes consist of rs5128and rs275646,there was no significant association between haplotype and simple obesity and central obesity.4.We used smoke,drink,hypertension,hyperlipidemia and diabetes as the environmental interactive factors to fit the model and found that among the haplotypes consist of rs2638360 and rs2933249,C-C haplotype and diabetes have positive interactive effect with simple obesity and central obesity?P<0.05?.Among the haplotypes consist of rs5128 and rs275646,C-T haplotype and hyperlipidemia have negative interactive effect with simple obesity and central obesity?P<0.05?.Conclusion1.Rs2638360 in AGTR1 gene was significantly associated with simple obesity and central obesity.2.There was a significant gene-gene interaction among the SNPs of AGTR1 gene and this interaction may influence the occurrence of simple obesity and central obesity.3.Haplotype C-T of AGTR1 gene increased the risk of simple obesity.4.Diabetes and hyperlipidemia may influence the occurrence of simple obesity and central obesity by the interaction with the two haplotypes of AGTR1 gene.
