Objective: To investigate the expression of SOX-2 and Ki-67 in colorectal adenoma,adenocarcinoma and normal intestinal mucosa,and to compare the relationship between their expression and proliferation.Methods: There were 120 specimens of colorectal adenoma,50 cases of adenocarcinoma and 50 cases of normal adenocarcinoma.The results showed that there were 50 cases of colorectal adenoma,Intestinal mucosal tissue in 20 cases.According to the degree of atypical hyperplasia,tubular adenomas were divided into two groups: low grade tubular adenoma group(n=30)and high grade tubular adenoma group(n=20).Adenocarcinoma was divided into three groups according to the degree of differentiation Differentiated adenocarcinoma(n=8),moderately differentiated adenocarcinoma(n=26),poorly differentiated adenocarcinoma(n=16).The expression of SOX-2 and Ki-67 in the specimens was detected by immunohistochemistry.Results:(1)SOX-2 and Ki-67 were not expressed or expressed weakly in normal intestinal mucosa,but expressed in colorectal tubular adenoma and adenocarcinoma;The positive expression rates in colorectal tubular adenoma were 22% and 30%,and were 70% and 78% in adenocarcinoma.;The expression of SOX-2 and Ki-67 in the three groups was statistically different.(2)In adenocarcinoma,the positive expression rate and expression level increased with the degree of malignancy,P < 0.05,with statistical significance;In tubular adenoma,the positive expression rate and expression level increased with the degree of dysplasia,P < 0.05,with statistical significance.(3)The expression of SOX-2 and Ki-67 was not correlated with sex,age,tumor size and lymph node metastasis in colorectal cancer patients.(4)Ki-67 and SOX-2 expression was positively correlated.Conclusion:(1)The positive expression of SOX-2 was enhanced with the increased of the degree of atypical hyperplasia in colorectal tubular adenoma and decreased differentiation of adenocarcinoma.(2)the positive expression of ki-67 was enhanced with the increased of the degree of atypical hyperplasia in colorectal tubularadenoma and decreased differentiation in adenocarcinoma.(3)The expression of Ki-67 were positively correlated with the SOX-2 expression,through the the joint detection of SOX-2 and Ki-67 could help determine the proliferation of colorectal tubular adenoma and adenocarcinoma,and provide a reliable theory basis for the early diagnosis,prognosis and treatment of colorectal cancer.
