Objective:By detecting respectively the interleukins 35 expression level via enzyme-linked immune sorbent assay(ELISA)in pleural effusion which come from patients with malignant pleural effusion(MPE),tuberculous pleural effusion(TPE)and Parapneumonic pleural effusion(PPE),to investigate preliminary whether it is meaningful in the differential diagnosis of MPE,TPE and PPE.By detecting IL-35 Through rt-pcr in carcinoma tissues and normal lung tissue adjacent to carcinoma tissues from non-small cell lung cancer(NSCLC)patients to analyze the expression of IL-35 in NSCLC tissue.By detecting IL-35 through ELISA in the serum from NSCLC patients and healthy persons to analysis the relationship between serum levels of IL-35 and clinicopathological features in NSCLC patients.Methods:1.By detecting the content of il-35 in PE by ELISA kit to analyze he expression of il-35 in MPE,TPE and PPE.2.By collecting cancerous specimens and adjacent normal lung specimens from 20 patients with NSCLC,and to detect the difference of il-35 expression level at m RNA level through rt-pcr.3,By measuring the expression of serum IL-35 in 78 non-small cell lung patients and 20 healthy persons by ELISA,to analyze whether there is any difference between the expression level of IL-35 in serum and the healthy group and lung cancer group,to reseach the relationship between IL-35 and clinical pathological features of NSCLC.Results:1.The content of IL-35 was the highest in MPE,the second in TPE,and the lowest in PPE.The content of IL-35 was higher in the pleural effusion with cancer cell than in the negative group.2.Rt-pcr results indicated that the expression level of il-35 in NSCLC patients was significantly higher than that of the normal lung specimens.In NSCLC patients,the expression level of il-35 in lung squamous cell carcinoma was higher than that of lung adenocarcinoma,which was statistically significant.3.Through detecting the IL-35 in the serum from the group of NSCLC patients and healthy persons group,the expression degree of IL-35 in NSCLC patients serum is far higher than the healthy persons.The expression degree of IL-35 in lung squamous cancer patients serum is higher than in the lung adenocarcinoma,but the difference was not statistically significant.The level of IL-35 in serum of NSCLC patients was detected by ELISA.It was found that there was no significant correlation between the serum IL-35 level in patients with NSCLC and the patient's gender,age,tumor tissue volume,However,there was a positive correlation between the serum IL-35 level in patients with NSCLC and tumor differentiation,TNM stage,and lymph node metastasis.Conclusions:Cytokine il-35 has a certain practical value for the identification of benign and malignant PE,and il-35 in cancer tissues has a certain significance in the classification of lung cancer.The expression level of il-35 in serum was positively correlated with NSCLC differentiation,TNM classification and lymph node metastasis,but was not significantly correlated with the gender,age,and primary tumor size of the patients.However,further large-scale and large data research are needed to discuss the determination.
