Objective: Macrophages activation is a critical event in immune response,varieties of stimulation in the microenvironment where macrophages reside would induce macrophages polarization toward M1 macrophage or M2 macrophage,thus elaborately regulating inflammatory response.The effect of HIV infection on macrophages activation remains unclear,there's study elucidating that long non-coding RNAs can regulate inflammatory response,our study aims at illuminating the effect of HIV infection on macrophages activation through researching the effect of HIV infection on the expression of lncRNAs of macrophages and the effect of lncRNAs on macrophages activation.Methods: Macrophages were cultured and stimulated with HIV-1,then macrophages were harvested after verification of successful HIV-1infection to macrophages.Gene chip was performed to test the expression of lncRNAs,the lncRNA that up-regulated the most after HIV-1 infection was chosen,an over-expression recombinant plasmid and a shRNA were designed and transfected into macrophages.The expression of MHC I,MHC II and M1 macrophage or M2 macrophage related molecules were detected by ELISA,WB and RT-PCR,thus researching the function of lncRNA in macrophages activation.Results: HIV-1 successfully infected the macrophages,and the high expression of lncRNA n266623 in macrophages after HIV-1 infection was verified through gene chip and RT-PCR.pcDNA3.1-lncRNA-n266623 and p Silencer1.0-U6-lncRNA-n266623 were obtained and transfected into macrophages,molecule markers of M1 macrophage such as IL-1?,IL-6,TNF-? and i NOS were down-regulated and molecule markers of M2 macrophage such as IL-10,CCL-18,Arg1 and Mrc1 were up-regulated in macrophages after transfection of pcDNA3.1-lncRNA-n266623,the expression of these molecule markers in macrophages after transfection of p Silencer1.0-U6-lncRNA-n266623 were of opposite tendency compared with above.The expression of MHC I and MHC II were down-regulated after transfection of pcDNA3.1-lncRNA-n266623 and up-regulated after transfection of p Silencer1.0-U6-lncRNA-n266623.Conclusions: The expression of lncRNA-n266623 was up-regulated in macrophages after HIV-1 infection,lncRNA-n266623 would down-regulate the expression of M1 macrophage related molecule markers and up-regulate the expression of M2 macrophage related molecule markers,which would induce macrophages toward M2 macrophage polarization and negatively regulate the immune response.
