Objective:To explore the clinical value of PD-1 in the development of non-small cell lung cancer(NSCLC)by detecting the expression of programmed death-1 receptor on CD3+ T lymphocytes in the lymph nodes of patients with non-small cell lung cancer(NSCLC).Methods: Fifty-one lymph nodes with NSCLC from January 2016 to December 2016 were collected.23 lymph nodes came from male patients and 28 lymph nodes came from female.The lymph nodes were divided into two parts,one of them were stained with hematoxylin-eosin(HE),the other of them were detected the expression of PD-1 on CD3+ T lymphocytes by flow cytometry(FCM).Results:1.Lymph node tissue specimens of patients with non-small cell lung cancer,metastatic lymph nodes in 26 cases,and 25 cases of non-metastatic lymph nodes.The percentage of PD-1 expression on CD3 + T cells was(50.08 ± 8.03)% in metastatic lymph nodes,which was higher than that in non-metastatic lymph nodes(36.25 ± 11.27)%,the difference was statistically significant(t = 5.208,P <0.001).2.The expression of PD-1 on CD3+ T lymphocytes in the lymph nodes in adenocarcinoma was significantly higher than that in squamous cell carcinoma(P <0.05).And the expression of PD-1 on CD3+ T lymphocytes in advanced stage lung cancer(stage III and IV)was significantly higher than that in early stage lung cancer(stage I and II).There was significant difference between the two groups(P <0.05).What's more,there was no clear correlation between PD-1 expression level on CD3+ T lymphocytes and the age?sex?smoking history(P> 0.05).Conclusion: 1.The expression of PD-1 on CD3 + T cells of metastatic lymph nodes in patients with non-small cell lung cancer was significantly higher than that in non-metastatic lymph nodes,which suggested that PD-1 and its ligands play an important role in the progression of non-small cell lung cancer.Which suggested that the PD-1 and its ligands were involved in the process of lymph node metastasis,and they were hoped to become an important indicator of tumor prognosis,and they could guide the immune treatment of lung cancer.2.The expression of PD-1 on CD3+ T cell in infiltrating lymph nodes of non-small cell lung cancer was significantly correlated with tumor type and staging of lung cancer,which suggested that PD-1 and its ligands were closely related to the formation,differentiation and progression of lung cancer,and they may provide new targets for lung cancer immunotherapy.
