Objective: This study aims to investigate the effect and mechanism of Honokiol on the proliferation,apoptosis,migration and invasion of human pancreatic cancer cell line SW1990.Methods: SW1990 were treated with Honokiol at concentrations of 0,5,10,20,30,40,80,120 ?mol/L for 24,48 and 72 h,respectively.CCK-8 assay was used to detect the cell proliferation.When the cells were treated with Honokiol at concentrations of 0,20,40,80?mol/L for 24 h,apoptotic rate of cell was detected by flow cytometry,cell migration ability was evaluated by wound healing,cell invasion ability was evaluated by tumour invasion assay,mRNA and protein expression of ?-catenin,C-myc,Survivin,Bax,Bcl-xl,Caspase-3,E-cadherin,Vimentin were detected by RT-PCR and Western blot,respectively.Results: Honokiol could significantly inhibited the growth of SW1990 cells in a time-dependent and concentration-dependent manner(P<0.05).The late stage apoptosis of SW1990 were respectively(2.1±0.44)%,(5.25±1.39)%,(25.75±1.00)%,(60.19±3.14)%.Apoptosis rate was positively correlated to Honokiol concentration(P<0.05).Honokiol inhibited metastasis and invasion of pancreatic cancer cells,the difference among the different concentration groups was statistically significant(P<0.05).The mRNA and protein expression of ?-catenin,C-myc,Survivin,Bcl-xl,Vimentin were down-regulated and Bax,Caspase-3,E-cadherin were up-regulated with concentration-dependence,which was detected by RT-PCR and Western blot.Conclusion: Honokiol could significantly inhibit the proliferation,metastasis,invasion and induce apoptosis of human pancreatic cancer cell SW1990.Honokiol could inhibit the migration and invasion of pancreatic cancer cells SW1990 by affecting the expression of ?-catenin,a key gene in the Wnt/?-catenin signaling pathway.
