Objective: To study the expression and significance of KDM1 A protein in nasopharyngeal carcinoma and its correlation with clinicopathologic features in nasopharyngeal carcinoma.Methods: The expression of KDM1 A were determined by the real time PCR,Western blot,and immunohistochemical staining.The chi-square test was used to exam the relationship between KDM1 A expression and clinicopathologic characteristics.The survival curves were estimated using the Kaplan-Meier method.The log-rank test was used to estimate the significance of the differences between the survival curves.A Cox proportional hazards analysis was performed to calculate the hazard ratio(HR)and the 95% confidence interval(CI)to evaluate the association between clinicopathologic characteristics and overall survival time(OS).Results:(1)Real time PCR and Western blot shown increased KDM1 A mRNA and protein expression in invasive nasopharyngeal carcinoma.And the positive rates of KDM1 A protein was 69.737%,which was significantly higher than that in adjacent mucosa tissues(6.667%)(p<0.05).(2)The positive rate of KDM1 A in patients with TNM3 stage were higher than that in early stage patient(p<0.05).(3)The positive stain of KDM1 A in nasopharyngeal carcinoma correlated with unfavorable prognosis.(4)Cox regression analysis shown KDM1 A positive expression was an independent prognostic factor.Conclusion:(1)KDM1A testing could predict the invasive behavior and higher clinical TNM stage of nasopharyngeal carcinoma;(2)KDM1A correlate with the advanced TNM stage of nasopharyngeal carcinoma and unfavorable prognosis.
