A Study On The Establishment Of Immune Thrombocytopenia Model Induced By Platelet GPIb? Antibodies

?Objective?To establish different kinds of ITP model induced by different anti-platelet membrane glycoprotein GPIb?a antibodies.?Methods?20 guinea pigs(6~8 week)were divided into 4 groups.5 guinea pigs in each group were intravenously injected with different dose of AN51(0.05 ?g/g,0.1 ?g/g,0.2 ?g/g)and 0.2 ?g/g IgG as control.The whole blood was collected from innerangular venous plexus.Platelets number was determined by an automated cell counter and Swiss-Jim method.Then,the similar protocol was used to establish ITP model of GPIb? humanized mice and nude mice by injecting with different concentrations of anti platelet GPIb? antibody SZ2 and R300,respectively.?Results?Venous injection 0.05 ?g/g ?0.1 ?g/g?0.2?g/g of AN51 decreased 0~5%?50%~60% ? 70%~80% platelets after 1/12 hours in guinea pigs group(P<0.01);intraperitoneal injection 0.05 ?g/g ?0.1 ?g/g?0.2 ?g/g ?0.4 ?g /g of SZ2 decreased 0~10%?20%~30%?50%~60%?80%~90% platelets after 6 hours in GPIb? humanized mice(P<0.01);intraperitoneal injection 0.05 ?g/g ?0.1 ?g/g?0.2 ?g/g R300 decreased 20%~30%?60%~70%?80%~90% platelets after 6 hours in nude mice(P<0.01).The nude mice,injected 0.2 ?g/g R300 everyday in 2 weeks,showed typical ITP clinical manifestations including large number of petechiaes or ecchymoses,on limbs,head and abdomen.?Conclusion?0.2 ?g/g AN51,0.4 ?g/g SZ2 and 0.2 ?g/g R300 could establish stable ITP model in guinea pigs,GPIb? humanized mice and nude mice respectively.R300(0.2?g/g)could establish typical ITP model in nude mice.
