Breast cancer is harmful to women' s health seriously,otherwise the traditional therapy is limited,so it is in need of immediately new tumor immunotherapy.Nowada-ys,that non-coding RNA in tumorigenesis mechanism research and appilication of the inhibitor targeted cancer therapy has becomes a big focus,in which the inhibitor of Z38 gene(antisense nucleic acid,si-RNA,etc.)has been shown to inhibit the proliferation of tumor cells by inhibiting Z3 8 to kill tumor cells effectively but not to normal cells.At the same time,with the PD-1/PD-L1 signaling pathway being elucidated,it has also become hot that the PD-1 or PD-L1 specific monoclonal antibody block the PD-1/PD-L1 negative regulatory signaling pathway to cure cancer.In this study,Z38 and PD-L1 gene were used as the research object to study their correlation in breast cancer.This study is aimed to investigate the molecular mechanism of Z38 by the clear molecular mechanism of PD-L1 in tumor.Since the new discovery of Z38,the research result of Z38's molecular mechanism is know little,confirmation of the relationship between PD-L1 and Z38 could help to study the mechanism of Z38.Through studying the Z38 and PD-L1-related pathway mechanism,it is maybe a good way to treat tumors with Z38 inhibitors to block PD-1/PD-L1 signaling pathway.This article specifically carried out as follows:Firstly,to pick up a breast cancer cell line as study material,we detected the expression levels of Z38 and PD-L1 in three breast cancer cells.Secondly,tumor cell' s proliferation rate was measured in metastatic cell lines that interfere with Z38 in breast cancer.Thirdly,we constructed and identified the metastatic cell lines that interfere with PD-L1 and detected the proliferation ability of these tumor cells,the killing ability of T lymphocytes to tumor cells and the proliferation activity of T lymphocytes.Lastly,we detect the exprssion of the other gene in metastatic cell lines of Z38 and PD-Ll,then investigated the relationship between Z38 and PD-L1 by RIP.Results as follows:At first,the relative expression of Z38 and PD-L1 was higher in breast cancer MDA-MB-231 cells.Secondly,the proliferation rate was decreased in breast cancer metastatic cell lines of interfering Z38.Thirdly,Thirdly,the lentivirus system and breast cancer metastatic cell lines of interfering PD-Llwas successfully constructed and the tumor cell proliferation activity of these tumor cells were decreased,the tumor killing ability of T lymphocytes was enhanced,and interfering strains promoted the proliferation of T lymphocytes.At last,the expression of PD-L1 was decreased in interfering Z38 cells and the expression of Z38 was also decreased in the interfering PD-L1 cell line.The RIP experiment directly demonstrates the relationship between PD-L1and Z38.In this study,we mainly obtained the stable MDA-MB-231 cell lines of interfering PD-L1 which could promote the proliferation of T lymphocytes.These results demonstrated the direct relationship between PD-L1 and Z38 by qPCR,Western Blot and RIP experiments.This work provides the basis for our future experiments.
