Oxidative stress plays an important role in the process of cerebral ischemia-reperfusion(CIR)injury.Therefore,flavonoids with strong antioxidant activity are effective in the treatment of cerebrovascular diseases such as CIR.The experimental research method of the system based on the pharmacology of Hawthorn Leaf Flavonoids in selected active molecule kaempferol(KA),and predict its anti CIR mechanism of injury,the protective effect of KA on animal models of middle cerebral artery occlusion in rats of CIR damage,E2 antioxidant pathway factor nuclear factor in brain tissue was detected by Real in Time-PCR related factor 2(Nrf 2)mRNA and heme oxygenase 1(HO-1)expression of mRNA,using the Western blotting method for determination of the expression of Nrf 2 and HO-1 protein,discussed the mechanism of CIR against damage of KA.1.According to the TCMSP database analysis and prediction,Kaempferol as one of the active ingredients in hawthorn leaves has the research significance,and its targets were closely linked with reperfusion,atherosclerosis and cerebral ischemia diseases,the Nrf2/HO-1 pathway can be considered one of the key mechanisms against CIR.2.CIR injury in rats was induced by middle cerebral artery occlusion,we studied the cerebral protective effect of long and short term prophylactic administration of kaempferol.Compared with IR group,kaempferol and hawthorn leaf flavonoids significantly reduced mortality,neurological score,infarct volume,and increased the antioxidant factor SOD content,decreased the peroxidation product MDA content,and the long-term administration kaempferol group had the best effect.The results suggested kaempferol could against CIR injury in rats,and the mechanism may be related to antioxidant.3.We investigated antioxidant mechanism of the neuroprotective effects of kaempferol combined with the systematic pharmacological analysis results,the results show that compared with the model group,kaempfetol increased the expression of Nrf2 mRNA and HO-1 mRNA,it also could promote Nrf2 transfer from cytoplasm to nucleus in brain tissue of CIR rats,and active the downstream HO-1 protein that have antioxidant effects.There was no significant difference among administration drug groups.Conclusions:System pharmacology was of great significance for the study of traditional Chinese medicine because of its accurate prediction results.Animal experiments prove that KA is the key component of hawthorn leaves flavonoids against CIR,Nrf 2/HO-1 is one of its antioxidant mechanism and effect of anti injury and preventive administration of mountain combined medication time correlated. |
