| Objective This study is one part of National Natural Science Foundation(NSFC)of China “Mechanism study of Tujia medicine Toddalia asiatica(L.)Lam.to inhibit the action of the RA based on on the mi RNA-146 a and TLR/NF-kappa B signaling pathway”(81360672),and Hubei Provincal Key Laboratory of Occurrence and Intervention of Rhumatic Diseases "Curative effect and mechanism study of Tujia medicines Toddalia asiatica(L.)Lam.on rheumatic arthritis"(OIR3007A).This study aimed to explore curative effect and mechanism study of Tujia medicines Toddalia asiatica(L.)Lam.on rheumatic arthritis.Methods Using improved karber method to carry out the Toddalia asiatica(L.)Lam.supplied in powder,water extraction,alcohol extraction of components and the ethyl acetate extract composition of acute oral toxicity studies,and is verified with SD rats;In the collagen induced arthritis model based on the given wind cold dampness,produced in conformity with the characteristics of traditional Chinese medicine the wind cold dampness of animal models of RA;In a laboratory of homemade three hundred gel for drug intervention,wantong the physique to stick as positive control,rats in normal group and model rats asck,evaluation of its general signs on RA rats,ankle weeks diameter,inflammation,grading,the effect of using flow cytometry technology,inspects the RA rat peripheral blood T lymphocyte subsets in the surface the effect of the content of specific antigen and Th1 / Th2 drift.Results(1)Toddalia asiatica(L.)Lam.all components,the maximum tolerance of 28.57 g/kg,median lethal dose of 3.464 g/kg,and supplied in the water extraction,alcohol extraction components and ethyl acetate extract components of the median lethal dose of 4.005 g/kg,3.196 g/kg and 2.901 g/kg,according to the various components of yield of conversion,that is equivalent to content were 49.40 g/kg,35.86 g/kg,30.98 g/kg.Under the same doses(28.57 g/kg)in mice mortality rate of ethyl acetate extract of the water extract,alcohol extract all components.Mouse lavage after Toddalia asiatica(L.)Lam.of different components of the main symptoms ofacute toxicity of be agitated,hyperactivity,oral,limbs and tail blue,salivate,bloodshot eyes tears appear even hunch over,shrink abdomen,rolling,died of respiratory failure,etc.(2)The wind cold dampness group is significantly higher than the CIA induction week of ankle joint diameter,peripheral blood immune factors TNF-alpha,IFNgamma induced group were significantly higher than that of normal group and the CIA,TNF alpha in synovial fluid is significantly higher than normal group and the CIA induced group;The CIA induced group and wind cold dampness of the femur and the ulna bone mineral density were significantly lower(P < 0.05),and bone tissue morphology also abnormal changes;Among them,the wind cold dampness of group of bone loss is more apparent,fracture and bone tissue morphological structure appear even distorted.(3)model group rats/CD3+,CD4+,CD8+/CD3+were significantly lower,three hundred rods gel group specific antigen on the surface of T lymphocytes in the peripheral blood of CD4+/CD3 +,CD8+/CD3+with model group and had significant difference before treatment,rise to normal levels;Rheumatoid arthritis(ra)model rats IFN-gamma/IL-4 imbalances,moving in the direction of proinflammatory factor;Toddalia asiatica(L.)Lam.gel topical treatment,the equilibrium is moving in the direction of inflammation suppression factor.Conclusion Toddalia asiatica(L.)Lam.oral medicine has certain acute toxicity,long-term use of liver and kidney function may cause serious damage,external use is a more safe and feasible for medicine;Toddalia asiatica(L.)Lam.topical treatment of rheumatoid arthritis has a certain effect,the treatment effect is by adjusting the T lymphocyte surface specific antigen expression and improve the realization of Th1/Th2 imbalance state. |
