Background and Objective:ATP citrate lyase(ACLY)is a key enzyme in the upstream of the lipid metabolism pathway.As researches show,that the expression and activity of ATP citrate lyase(ACLY)are increased in some cancers.Our previous studies have confirmed that the high expression of ACLY is associated with advanced gastric cancer and poor prognosis.However,the significance of ACLY in colon cancer is infrequently reported.This study aims to explore the expression of ACLY in colon cancer and its clinical pathological significance,in order to provide new ideas for the diagnosis and treatment of colon cancer.Method:1.These 50 patients with colon cancer confirmed by pathology after operation in Zhong Da Hospital affiliated to Southeast University during 2015.08-2016.06 are selected as subjects.The on necrotic area of tumor and the tissues adjacent to the edge of cancer 5 cm above are taken as samples.50 cases of colon cancer specimens are the study group,and the corresponding adjacent tissues are the control group.2.The relative expression of ACLY is detected by immunohistochemical method.The positive staining of ACLY is performed by semi quantitative analysis.3.Arrange the patient's name,age,preoperative diagnosis and treatment,operation time,pathological number,pathological data(WHO classification method)and the ACLY results.4.Using SPSS 17.0 software,?2 test for statistical analysis of the experimental data,p>0.05 statistically significant.Result:1.The results of IHC show that ACLY is mainly localized in cytoplasm and cell membrane.In the 50 colon cancer tissues,there are 40 cases with the high expression of ACLY,and 10 cases with the low expression of ACLY.The expression of ACLY in colon cancer is strong.In the 50 adjacent tissue,there are 8 cases with the high expression of ACLY,and 42 cases with the low expression of ACLY.The expression of ACLY in adjacent tissue is week.Compared with adjacent tissue,the expression of ACLY in colon cancer tissue is increased significantly(p=0).2.Analyzing the expression of ACLY in colon cancer tissue and clinical pathological parameters,we find that high expression of ACLY is related to colon cancer invasion,lymph node metastasis and cancer TNM stage(p=0,0.005,0.007).But the age,gender,tumor size,histology grade had no significant effect with ACLY(p>0.05).Conclusion:ACLY is highly expressed in colon cancer tissues and associated with disease progression,which can be used as a clinical marker for evaluating the staging of colon cancer.ACLY may be a new target for the diagnosis and treatment of colon cancer.
