Objective:In order to provide experimental basis for further experimental study and clinical application,the effects of Kangangxin on healing of cartilage defect of knee joint of New Zealand rabbits were observed,and the possible mechanism of repair of cartilage defect was preliminarily discussed.Through the establishment of in vitro cultivation of KOA cartilage cell line and observe rehabilitation new fluid influence on KOA cartilage cell proliferation,and the determination of recovery in the new liquid under the action of KOA rabbit chondrocytes P44/42 mapk protein expression.Experimental methods:1.Experimental study on the repair of cartilage defect of rabbit knee joint by rehabilitation new fluid.The weight 3.5 4.5 kg of clean level,male New Zealand rabbits,randomly divided into four groups,new liquid high dose group,model control group,rehabilitation rehabilitation in new liquid dosage group,the rehabilitation of new liquid low dose group,each group of 18(6 points three times,each time period only).The defect model of the femoral medial condyle of the right knee joint of the rabbit was induced by surgical drilling method with 3% pentobarbital sodium(1ml/kg).After the model was established,each group was injected with different concentrations of convalescent fluid at the bone and joint defects.In the bone defect of 4,8,12 weeks to groups of animals,observe the specimen and to HE staining and Masson staining of tissue slices,according to the results of the dye image analysis judgment recovered new liquid of rabbit bone defect repair status and the role of organizational change.2.Experimental study on the proliferation of chondrocytes and p44/42mapk protein expression in rabbit KOA.The clean grade of 3.5-4.5kg and the male New Zealand rabbit were anesthetized with 3% pentobarbital sodium(1m L/ kg)via ear vein.The modified articular mark method was used to reproduce the KOA model of rabbits.After the operation,the animals were driven away for 30 minutes daily,and the KOA model was successfully duplicated at the 4th week.After the New Zealand rabbits were killed by air embolization,the rabbit KOA chondrocytes were established by enzyme digestion of primary cell culture.The expression of p44/42 MAPK protein in rabbit KOA chondrocytes was detected by Western blotting.Experimental rehabilitation new fluid.4 weeks after surgery,the model control group: the cartilage defect was obvious and the granulation tissue was visible.In the high,medium and low dose groups of the convalescent new fluid,all the defects were obvious and the white bone tissue was visible.Eight weeks after surgery,the fiber-like tissue was seen at the bottom of the defect.Low dose group of rehabilitation new fluid: filled cartilage in the defect area;The dose group in the new fluid of rehabilitation: the defect area can be filled with cartilage and luster;High dose group:defects in the defect area are not obvious,filling the cartilage is smooth,there is a small amount of osteophyte formation.In the 12 weeks after surgery,fibroid tissue was seen in the defect area of the model control group.Low dose group: the defect area was filled and the surface was rough.The dose group in the new fluid of rehabilitation: the defect area was filled up and the surface was dull.High dose group: articular cartilage structure is good,the defect area is filled,the surface is smooth,and the boundary is blurred with normal area.1.2After HE staining in the cartilage defect,the results of the semi-quantitative scoring were obtained.Postoperative 4 weeks,8 weeks,12 weeks three time points results show that: compared with model control group,new liquid recovery high,medium and low dose group of semi-quantitative score were lower,the difference had statistical significance(P < 0.05).1.3Masson staining results were obtained in the 12 weeks after surgery.Model control group: cellulose was generated but arranged in disorder,and cartilage cells secreted little collagen fibers.The low dose group of rehabilitation new liquid: the structure of the repair place is more disorganized,and the cartilage cells secrete less collagen fibers.The dose group in the new fluid of rehabilitation: there were more chondrocytes,but the secretion of collagen fibers was less,and the cellulose was more and arranged relatively regularly.The high dose group of rehabilitation new liquid: the structure of the repair place is arranged in a regular pattern,the cartilage cells are more and more orderly,the fiber content is obviously decreased,and the collagen fiber content is much.3.Experimental study on the proliferation of chondrocytes and p44/42 mapk protein expression in rabbit KOA.Compared with the model group,10 ng / m LL 20 ng / m LL + 40 ng / m LL + 80 ng /m L rehabilitation group could promote the proliferation of rabbit KOA chondrocytes(P < 0.05),and the higher the concentration,the stronger the proliferative effect,and the lower the expression level of p44r42 protein was in the 10 ng / m L20 ngm L20ng-1/ m L rehabilitation group compared with the KOA group.The difference of 40ng/m L rehabilitation group was statistically significant(P < 0.05),and the higher the concentration was,the stronger the inhibitory effect was.conclusion1.The recovery of new fluid has a certain effect on the cartilage defect of the rabbit's knee joint.2.Rehabilitation can promote the proliferation of KOA chondrocytes,decrease the expression of p44/42 mapk protein in MAPKs signaling pathway,play a role of anticartilage degeneration and promote bone repair.
