Objective:To observe the effect of total flavonoids of Rhizoma Drynariae on os teogenic factors induced by long segmental bone defect in rats,and to provide a new method and new basis for clinical treatment of long segmental bone defect s.Methods:Four groups of SD rats were randomly divided into four groups: grou p A,group B,low dose group,group C middle dose group and group D.All rats were made with long tibial long bone defect model.The lower edge of the knee 0.5cm at the tibial tubercle about a length of about 1.5cm longitudinal in cision,layer by layer to open the fascia,the separation of muscle,completely e xposed tibial tubercle under the bone,hand surgery jaw truncated tibia,Interce ption of about 6mm bone,the tibial intramedullary fixation device from the prox imal end of the bone defect inserted into the tibia,proximal fixation after the fi xed distal,fixed firmly,antibiotic bone cement stuffing.Bone cement is complet ely hardened after suturing fascia,skin incision.Postoperative administration of penicillin sodium 80,000 U / intraperitoneal injection,continuous injection for 3days.The rats in the low-dose group were treated with distilled water in the co ntrol group and low in the low dose group.The total flavonoids in the middle dose group were given the total flavonoids of Rhizoma Drynariae(202.5mg /(k g.d))The rats in the dose group were given the total flavonoids of Rhizoma Dr ynariae 607.5 mg /(kg.d)for 6 weeks.After the model was established,the ex pression of osteogenic factors was detected by ELISA,and the morphology of th e induced membrane was observed by histopathology.Application of SPSS20.0 sof tware for statistical analysis of data.Results:Rats after anesthesia awake can be free in the cage,can stand feeding,left limb mild limp,3 days after the limp symptoms disappeared.Wound healing well,no rednessexudation,skin surface suture 7 days or so natural fall off,incision are Class A healing.The rats in the blank control group were killed by anesthesia over 1 week after the operation.The low dose group died in the high dose group.No deaths were found in the group.In the course of the experiment,except rats,the rats were all in good condition,the hair was neat and shiny.3 days Afterothe operation,the rats were obviously resistant,responsive,rapid,Normal,no diarrhea and other abnormalities.In the whole experiment,no abnormal nose and mouth exudate.Each rat weight was weighed every 2 weeks.All rats survived until the experiment was completed.X-ray performance can be seen on the left tibial long segment of bone defects,bone defects within the high-density material stuffing,tibial visible intramedullary fixation device fixed.The expression of OD value of BMP-2 positive expression was blank control group<low dose group <middle dose group <high dose group.Compared with the high dose group,P = 0.000,the difference was statistically significant in the blank control group,low dose group,middle dose group and high dose group.The positive expression of VEGF was similar to that of BMP-2,and it was the control group <low dose group<middle dose group> high dose group.Compared with the high dose group,P = 0.000,the difference was statistically significant in the blank control group,low dose group,middle dose group and high dose group.Pathological observation of the blank control of the composition of fibrous cells arranged sparse,clear structure,containing a small amount of microvessels.Low dose group: fibroblasts are arranged sparsely distributed,structurally clear,containing a small amount of microvessels.The dose distribution of fibrous cells in the distribution of the rules,dense,clear structure,rich in microvessels.High-dose fibroblasts are arranged in a regular,dense,clear structure,rich in microvessels.Conclusions:.The total flavonoids can promote the expression of osteogenic fact ors in the membrane,and promote the healing of bone,which is beneficial to t he transportation of minerals such as calcium,promote the deposition of calcium salt in the bone,accelerate the formation and calcification of new bone and sh orten the time of osteogenesis.
