Objective:Using the streptozotocin to establish diabetes rat model,observe the effect of The total anthraquinones in Polygonum cuspidatum and Panax Notoginseng Saponins on diabetic rats.Different groups of rats were detected results,include blood glucose,body weight and urinary albumin,renal tissue superoxide dismutase(SOD),malondialdehyde(MDA),total antioxidant capacity(T-AOC)level,and bone morphogenetic protein(BMP-7),connective tissue growth factor(CTGF),platelet derived growth factor(PDGF-BB),glomerular vascular endothelial growth factor(VEGF)protein expression.To research the effects of combined use The total anthraquinones in Polygonum cuspidatum and Panax Notoginseng Saponins on oxidative stress and pathological changes in diabetic nephropathy,and to explore the mechanism of action of diabetes mellitus and its related complications.Methods: By intraperitoneal injection of STZ in normal healthy SD rats to establish the model of diabetes,induced citric acid sodium citrate buffer solution was prepared with streptozotocin STZ(65mg/kg),blood glucose values greater than 16.7mmol/L for three days can treat as the successful modeling.The model rats were randomly divided into groups,model group,Yuquan pill group,Total Anthraquinone of Polygonum cuspidatum group,Panax Notoginseng Saponins group,Panax Notoginseng Saponins combined with Total Anthraquinone of Polygonum cuspidatum group(Combination group).The model group and the blank group were given the same amount of normal saline every day,the drug group were given the corresponding dose of drugs,continuous administration for 8 weeks.24 hour urine was collected from the metabolic cage at the end of the eighth week after the model was used to determine the content of urinary microalbumin.After intraperitoneal anesthesia,the rat kidneys were removed,and the right kidney to prepare the kidney tissue homogenate,respectively,SOD activity,MDA content and T-AOC content were determined.The left kidney using hematoxylin eosin staining to observe the renal pathological changes,and using the streptavidin biotin complex method of biological immune group,the expression was observed in renal cortex area of glomerular and renal tubular growth factor protein.Using optical microscope,10*40 field of view were observed.The distribution and staining of positive products were observed under 25 visual fields randomly selected from each group,take photos.Analyzing the optical density of the positive target in each field of vision,using integral optical density(IOD)to represent the quantity of the index,and compare the integral light density.SPSS19.0 software was used to analyze the mean of each group.Results: 1 model group: STZ induced diabetic rat model was successfully replicated.After intraperitoneal injection of STZ,blood glucose levels were significantly increased,Blood glucose value greater than 16.7mmol/L.After modeling,the animals showed the general morphological characteristics of diabetes mellitus,and the amount of drinking water and urine volume were significantly increased,and the color and the activity were worse.In the model group,the urine albumin content increased significantly in 24 hours.Pathological changes showed that the glomerular atrophy and edema were severe in the model group,and there were edema,proximal convoluted tubule and proteinuria.The renal capsule narrows,and the glomeruli diffuse.Immunohistochemical staining revealed.The expression of the model group was significantly higher than that of the blank group at cytokines associated with the diabetic nephropathy.2 Administration group:In the body weight,blood glucose levels remained stable,and blood sugar values decreased.Urinary albumin content was lower than that of model group.The pathological changes showed mild edema,and the edema of proximal convoluted tubule was relieved and the renal capsule was narrowed.Each group had a protective effect on diabetic kidney tissue.Immunohistochemical staining result showed that the expression levels of cytokines were decreased in each administration group.3 urinary albumin excretion rate in the combined group was significantly lower than that in the Total Anthraquinone group,PNS group in 24 hours.Combined group was more effective to reduce protein excretion than Total Anthraquinone group and PNS group.Conclusion: The total anthraquinones in Polygonum cuspidatum and PNS has protective effect on renal tissue of diabetic nephropathy.It has a good effect in controlling blood sugar,reducing microalbuminuria,anti oxidative stress and reducing the pathological changes of renal tissue.The effect of combination therapy on reducing urinary albumin excretion was more obvious than that of single drug.Anthraquinone combined with Panax Notoginsen can be used as an effective material basis for the prevention and treatment of diabetic nephropathy.
