Objective: To observe the effects of total flavonoids of peony flower on repeated cerebral ischemia reperfusion in mice,and the protective effects of focal cerebral ischemia reperfusion in rats,effects of on inflammatory cascade response and brain energy metabolism in rats with focal cerebral ischemia reperfusion,and the effect of laser Doppler on the circulation of blood in four experimental studies.Discuss the effects of the The peony flower total flavonoids on cerebral ischemia reperfusion injury and its protective mechanism.Methods: Repeated cerebral ischemia reperfusion model in mice,bilateral common carotid artery ligation to block brain tissue blood supply 15 min,then loosen the artery clamp to restore the perfusion 15 min,and finally,the bilateral common carotid arteries were ligated to block the blood supply 15 min.In sham operation group,only the separation of carotid artery but without ligation.Each drug group was given the corresponding drugs,and once a day continuous administration for 7 days,Sham operation group and ischemia reperfusion group(model group)were given the same volume 0.5% CMC.In 7th days,after intragastric administration of 1h,the model was established,and after reperfusion for22 h,to determine the NSE level in the serum of mice,and determination of the IL-1??IL-6 ? IL-10 in brain homogenate and the pathological changes of brain tissue were observed by HE staining.The nutritional and protective effects of A on focal cerebral ischemia reperfusion in rats.Methods The focal cerebral ischemia and reperfusion model was made by thread embolism of MCA.That is,the middle cerebral artery occlusion by thread embolism,and reperfusion after 2h.In sham operation group,the carotid artery was separated,and the suture was not treated.Positive group(nimodipine group and NaoLuo Tong group),total flavonoids of peony flower large,medium and small dose group corresponding drugs were given,once a day,continuous administration for 5 days.Sham operation group and ischemia reperfusion group(model group)were given the same volume 0.5% CMC.At 5thdays,the model was established after administration of 1h,and after reperfusion 22 h,the neurological function of rats was evaluated,determination of NSE,EPO level in serum,and the levels of GDNF and TGF-? were measured by brain homogenate,the percentage of infarct volume was calculated by TTC staining,HE staining was used to observe the cerebral cortex and hippocampus in rats,and the positive expression of ICAM-1 and NGF were detected by immunohistochemistry.Effects of Total flavonoids of peony flower on focal cerebral ischemia reperfusion in rats,the modeling and drug delivery methods are the same as experiment two.After 22 hours of reperfusion,the blood was taken from rats,and determination of MPO,SOD and MDA levels in the serum,brain tissue was used to make 10% brain homogenate,and determinate the TNF-?,IL-1?,IL-6,ATP,VCAM-1,Asp,Glu and the test related indexes of Coomassie brilliant blue.Dynamic observation of the effect of total flavonoids of peony flower on cerebral microcirculation in mice by laser Doppler blood flow dynamic monitoring system(PERIMED,PF5001).The male and female are half,and positive group(nimodipine group and Nao Luo Tong group),total flavonoids of peony flower large,medium and small dose group corresponding drugs were given.Once a day and continuous administration of 10 d.In 10 th days,after 10 d administration of 1h,anesthetized mice.The probe was inserted into the base and fixed on the right side of the skull with biological glue.After the blood flow was stable,the bilateral carotid artery ligation was performed to block the blood supply,to record the average blood flow decreased the percentage.Results: Making repeated cerebral ischemia reperfusion in mice model successfully.The results of the experiments show that the total flavonoids of peony flower can reduce the NSE level in serum,and reduce the brain tissue homogenates of IL-1? and IL-6 level and improve the content of anti-inflammatory factor IL-10.It can obviously improve the ischemic injury of cerebral cortex.Making MCAO model successfully.The results of the experiments show that total flavonoids of peony flower can effectively improve the nerve function of rats,reduce the mortality rate,the down-regulation of NSE and the level of ICAM-1,upregulation of GDNF,NGF,TGF-? levels or expression,and the upregulation of hypoxia factor EPO level,improve the cortex and hippocampus of rat brain injury.Making MCAO model successfully.The results of the experiments show that the total flavonoids of peony flower can reduce the content of MDA in serum and MPO in brain tissue,upregulation of SOD activity in brain tissue,decreased significantly in the braintissue of TNF-?,IL-1?,IL-6 and VCAM-1 levels,decreased the content of Asp,Glu,and improve the energy metabolism of the brain function,different degree of upregulation of ATP of each type of water level.The results showed that the total flavonoids of peony flower could promote the microcirculation of the brain tissue of mice,and could antagonize the decrease of the blood flow of the bilateral common carotid artery.Conclusion: Experimental study on the effects of total flavonoids of peony flower on the mouse model of repeated ischemia reperfusion,the model of focal cerebral ischemia in rats,and the laser Doppler detection: Total flavonoids of peony flower can reduce free radicals and reduce the content of excitatory amino acids,inhibiting the inflammatory cascade,improve brain energy metabolism,promote the release of neurons and nerve system of nutrition and growth factors,improve the pathological injury of cerebral infarction and brain tissue,reduce the brain injury caused by ischemia reperfusion,to protect the brain organization.
