Objective: To explore the effects of triethylamine subacute oral administration on body weight and liver and kidney of male rats,provide the basis for a comprehensive evaluation of the toxicity of trimethylamine.Methods: The male SD rats were randomly divided into three experimental groups and one control group with 8rats in each group.The experimental groups were administrated with triethylamine one time daily for 28 days and the doses were 25mg/kg?50mg/kg?100mg/kg of trimethylamine,the rats were weighed once every seven days,quantitative feeding,calculate the rat food intake and food utilization.At the end of the exposure,animals were sacrificed to measure serum aspartate aminotransferase,alanine aminotransferase,alkaline phosphatase and globulin,albumin total protein and creatinine,uric acid and urea nitrogen,weighed heart,liver,testis,lung,kidney organ weight,organ coefficient calculation,pathological examination was performed for liver and kidney.Results:(1)The general situation of experimental animals:compared with the control group,the experimental animals in each dose group were slow to grow,decreased activity,rough hair,increased nasal secretions and more sensitive to external stimuli,increased with the dose and time of exposure,and high-dose group of animals were more intense response to the operation,with aggressive behavior.Then there were no abnormalities in the control group.(2)The effects of triethylamine on the body weight of experimental animals: there was no significant difference in body weight between the first week and the second week after exposure.At the end of the 3rd week and the 4th week,the body weight of the rats in the three dose groups decreased,compared with the control group,the difference was statistically significant(P <0.05).At the end of the 4th week,compared with the bodyweight of the middle and low dose groups,the high does groups rats were also statistically decreased(P <0.05),and with the increase of the exposed dose and time,the more obvious the decrease of body weight,and there was a significant dose—response relationship.(3)The quality of viscera and the number of viscera in the experimental animals were significantly higher than those in the control group(P <0.05).There were no significant differences in the quality and organ coefficient between the heart,the testis and the lung in the control groups.(4)After 4 weeks of exposure,the contents of aspartate aminotransferase and alkaline phosphatase in middle and high dose groups were higher than those in the control group and the control group and the low dose group,the difference was statistically significant(P <0.05).The total protein,albumin and globulin were significantly higher in the high dose group(P <0.05),and the difference was statistically significant(P <0.05).LDL and HDL in the high dose group were significantly lower than those in the control group(P <0.05).The levels of triglyceride,low density lipoprotein and high density lipoprotein in the high dose group were significantly lower than those in the control group(P <0.05).The total cholesterol(CHO)decreased in the middle and high dose groups,and the difference was statistically significant(P <0.05)compared with the control group.UA and BUN in the low,medium and high dose groups of triethylamine were significantly higher than those in the control group(P <0.05).(5)Pathological examination of liver and kidney of experimental animals:under the microscope,the structure of the liver lobules in the control group was clear,the structure of the liver cells was intact,the cytoplasm was filled,the nuclei were normal and the arrangement was uniform and no abnormal changes were observed.Compared with the control groups,the low dose groups showed mild hepatic edema Cytoplasm loose and other phenomena,the rest did not see abnormal changes.In the middle and the dose group experimental animals,the liver tissue structure was blurred,hepatocyte edema,necrosis and cytoplasm were balloon-like,nuclear atrophy and lysis,and the high dose group was more obvious.In the control group,the renal tissue structure was clear and complete,the glomerular volume was normal,the renal tubular structure was complete,no epithelial cell shedding,no inflammatory cell infiltration,there were no abnormal change.Compared with the control group,the low dose group showed increased glomerular volume,no inflammatory cell infiltration,tubular swelling and expansion,no epithelial cell shedding;middle dose group glomerular volume increased,no inflammatory cell infiltration,renal tubular swelling and expansion,and epithelial cells fall off;high dose group of kidney bleeding,glomerular volume was significantly increased,and inflammatory cell infiltration,tubular expansion and expansion,epithelial cells were shedding.Compared with the control group,the liver and kidney tissues of the middle and high dose groups were statistically significant(P<0.05).Conclusion:(1)Triethylamine can inhibit the weight gain of rats,and with the increase of dose and exposure time the weight loss is more obvious,and there was an obvious dose – response relationship.(2)Triethylamine exposure can lead the rat liver and kidney organ coefficient increased,liver and kidney blood biochemical indicators abnormalities,and with the increase in dose increased,indicating that triethylamine induced liver and kidney function damage in rats.(3)Triethylamine exposure induced rat liver cells edema,degeneration,necrosis and renal congestion,renal tubular swelling,interstitial inflammatory cell infiltration and glomerular volume increased,indicating that triethylamine can cause rat liver and kidney organic damage.
