Objective:To study the expression and significance of microRNA-214/218(miR-214/218)and tight junction protein Claudin-2 in acute kidney injury mice model at the different time points following ischemic/reperfusion.Methods: Male C57BL/6J mice as the research object,randomly divided into three groups:the control group(C group),sham operation group(S group)and the ischemic/reperfusion group(IR group).The S group and IR group were divided into six sub-groups respectively according to the reperfusion time point.Using Real-time fluorescent quantitative(qRT-PCR)to detect the expression of mi R-214/218 and Claudin-2 mRNA in renal tissue;Western Blot to test Claudin-2 protein expression level;Immunohistochemical staining to detect Claudin-2 distribution in renal tissue;HE staining observation renal pathological damage;Renal pathological score accessed pathological damage.Results:IR group after reperfusion kidney pathological damage grade level increased gradually,24 h was the most serious injury,the pathological damage score was 3.30±0.130,after gradually returned,C and S group had no signicance pathological damage,IR group and group S point at the same time the renal pathological damage of tubulointerstitium differences had statistical significance(P<0.05);Claudin-2 mRNA and protein expression in IR group gradually reduced after reperfusion,to 24 h reperfusion Claudin-2mRNA expression level the lowest was 3.85 times lower than the baseline values,Claudin-2 expression level relative to 24 h reperfusion was 0.30 ± 0.031,then gradually returned,the Claudin-2mRNA expression level differences had statistical significance of the same timepoint between IR group and S group(P<0.05);IR group in 0h-5d the Claudin-2 protein expression of the distribution difference was statistically significant(P<0.05);miR-214/218 after kidney ischemia-reperfusion expression gradually rised,24 h to the peak,respectively increased 7.42 times and 12.92 times than the baseline,and then gradually backed to baseline,group C and S miR-214/218 expression level continued low.Conclusions: miR-214/218 and Claudin-2 protein expression level changes associated with kidney damage after ischemia reperfusion,but the specific regulating relations between mi R-214/218 and Claudin-2 still need to further research.
