The rising morbidity and mortality of cancer significantly threaten human health,and the complexity and heterogeneity of cancer carry with formidable treatment challenges.In recent years,the rapid development of sequencing technology brings unprecedented opportunities for precision medicine and a large amount of precision medicine knowledge have accumulated.However,there is still a long way to go for doctors to utilize these knowledge in clinical practice according to omics data of patients.Searching systems of precision medication which can be conveniently accessed by patients and doctors are waiting to be developed.In this paper,we established a searching system for cancer precision medication.Firstly,we used a standard analysis workflow to call traits from transciptome data,including single nucleotide variants,gene fusions,alternative splicing events and gene expression levels.Secondly,we matched the precision medicine knowledge in our knowledgebase with these omic traits and made relevent precision medication recommendations.This searching system contains three databases and one searching algorithm:Patient Database,which stores the omic traits of patients;Precision Medicine Knowledgebase,which stores the integrative data from four precision medication resources,including My Cancer Genome,National Comprehensive Cancer Network guildlines,companion diagnosis in the drug labels of U.S.Food and Drug Administration,and predictive models of drug responses based on Genomics of Drug Sensitivity in Cancer;Reference Database,which stores reference and extended information to assist in the searching process;Searching Algorithm,which is used to link the three databases and implement medication strategies and searching rules.In the application based on transcriptome data from gastric cancer patients,our searching system reported ten potential precision medicine options,such as imatinib and so on.
