Objective:To investigate the mechanism of curcumin induced autophagy in cervical cancer HeLa cells via mTOR/eEF-2K pathway.Methods:1.With different concentrations and different time of curcumin on cervical cancer HeLa cells,cell proliferation was detected by MTT method.2.The experiment was divided into four groups: blank control group,curcumin group,mTOR activator MHY1485 group,curcumin + MHY1485 group.Western bolt method was used to detected to autophagy related protein LC3-II/LC3-I,p-mTOR,eEF-2K protein expression.Results:1.Curcumin had evidently inhibition to the cervical cancer HeLa cells with dose-dependence and time-dependence.When the inhibition of cell proliferation reached 50%,the concentration and time as the experimental concentration and time,that is,30 μmol/L and 48 h.2.Compared with the control group,the expression of LC3-II/LC3-I increased significantly in the treatment group(P<0.05);MHY1485 group decreased(P<0.05).Compared with the curcumin group,the expression ofLC3-II/LC3-I was decreased significantly in curcumin +MHY1485 group(P <0.05).3.Compared with the blank control group,the expression of p-mTOR and eEF-2K was significantly decreased in curcumin group(P<0.05);the expression of p-mTOR and eEF-2K increased in the MHY1485 group(P<0.05);compared with the curcumin group,the expression of p-mTOR and eEF-2K increased significantly in curcumin + MHY1485 group(P<0.05).Conclutions:1.Curcumin could inhibit the proliferation of cervical cancer HeLa cells.2.MTOR/eEF-2K signaling pathway may be involved in the induction of cervical cancer HeLa cells by curcumin.
