Objective To investigate the association between cyclooxygenase-2 gene polymorphisms and susceptibility in Parkinson's disease.Methods Case-control study was adopted in this clinical study.A total of 52 patients with Parkinson's disease were recruited as the case group,who were enrolled in the neurology clinic or inpatients in the department of neurology in The Affilicated BenQ Hospital of Nanjing Medical University during January 2016 to February 2017.Over the same period,60 cases of non Parkinson's disease matched by age and sex were involved as the control group,who were selected from the medical examination center of BenQ Hospital by means of simple random sampling method.The blood samples of both the case group and the control group were extracted with deoxyribonucleic acid.Polymerase chain reaction was used to amplify the target gene.By the method of restriction fragment length polymorphism,4 functional single nucleotide polymorphisms(-765G>C?-1195G>A?-1290G>A and 8473T>C)of cyclooxygenase-2 gene were detected respectively.Then we recognized which genetype of cyclooxygenase-2 gene each sample belonged to in the gel imaging system.?2 test was applied to evaluate the association between different genotypes and alleles of cyclooxygenase-2 gene and susceptibility in Parkinson's disease.Results There seemed no significant difference in functional single nucleotide polymorphisms sites-765G>C,-1290G>A and 8473T>C of cyclooxygenase-2 gene when comparing the genotype frequencies and allele frequencies between the case group and the control group(P>0.05).The different distribution of genotype frequencies in the-1195G>A single nucleotide polymorphism site of cyclooxygenase-2 gene made significant sense in both groups(P<0.05).However,this difference became no statistical significance when it came to the distribution of alleles G and A between the two groups(P>0.05)?Conclusion We found that in the Han population of East China,the single nucleotide polymorphisms sites-765G>C,-1290G>A and 8473T>C in cyclooxygenase-2 gene have no relations with the morbidity of Parkinson's disease.And the-1195G>A single nucleotide polymorphism site of cyclooxygenase-2 gene may be related to the susceptibility of Parkinson's disease,and further studies should be carried out to confirm it.
