Liver cancer is a serious therioma, which is commonly treatment by surgical resection, radiation therapy, immunity therapy, chinese medicine treatment. But the clinical treatment effect is not very good There are many molecular imaging technologies to diagnose diseases in clinical diagnosis, such as optical imaging, magnetic resonance imaging, computed tomography(CT)and so on.CT with high resolution and low sensitivity is difficult to apply in low density soft tissue imaging. While MRI is sensitivity to apply in soft tissue imaging with high temporal and spatial resolution and high sensitivity. Therefore, biocompatible multifunctional nanomaterials with its function are powerful tools to achieve multimodal imaging. We design a multifunctional nanoprobe to diagnose early liver cancer. The nanoprobe was conjuncted with aptamers which are bioactive molecules by surface chemical modification technology.Detail are described as below:(1)The multifunctional nanoprobes of Fe3O4 core with fluorescent SiO2 shell are synthesized by a facile improved Stober method.Through changing the amount of TEOS and APTES to regulate the thickness of the silica shell,we design three different sizes of molecule targeted nanoprobes(S-MFS,M-MFS and L-MFS).(2)The three different sizes of nanoprobes were evaluated by its chemical properties, cytotoxicity and targeting. The magnetic fluorescent nano biological probes, which are magnetic, fluorescent and targeted, have good dispersibility, stable fluorescent property, low cytotoxicity and good biocompatibility. The nanoprobe was selected for further use which is with good dispersion, stable fluorescence properties, low cytotoxicity and good biocompatibility.(3)The nanoprobe with good performance can be used for early diagnosis of the tumor both in cell imaging and MRI imaging in vivo and vitro. It is an effective measures to monitor in situ of tumour with more perfect disease evaluation index.
