| The Curcuma wangsiensis S.G.Lee&C.F.Liang(CW)and Curcuma aromatica Salisb(CA)are belonged to Curcuma(Zingiberaceae)as herbaceous perennial,their rhizome were recorded to Chinese Pharmacopoeia as the commonly used Chinese medicine,and having resolved depression,qi and the effect of dispelling blood stasis.Previous study of chemical constituents showed their is rich in curcuminoids and volatile oils,similar to those in Curcuma longa L.Modern pharmacological studies have shown that curcumin was isolated from Curcumca longa L has remarkable antioxidative activity and neuroprotective effects.To further find the active ingredient in this genus,cell model by H2O2-induced was established in our study.Antioxidative activity was evaluated for extracts of CW and CA 95%EtOH extract.including(high concentration of EtOH extract,HCE),75%EtOH extract(moderate concentration of EtOH extract,MCE),methanol extract(ME),dichloromethane extract(DE)and petroleum ether extract(PE),and elucidate preliminarily their mechanisms.The present findings indicated that the five kinds of extracts(MCECW,HCECW,DECW,MCECA,HCECA)reveal anti-oxidative effects against H2O2-injured PC12 cell.Their effects were decreased the lactate dehydrogenase(LDH)release and level of reactive oxygen species(ROS),attenuated the intracellular Ca2+ overloading,increased cell apoptotic rate and antioxidant enzyme system(SOD.CAT)and antioxidants(GSH)content,stabilized mitochondrial membrane potential(MMP)and enhanced antioxidative ability,maintained mitochondrial membrane integrity.stabilized Ca2+content,up-regulated the Bcl-2 protein level,down-regulated the Bax and Caspase-3 protein expression to inhibition of apoptosis.This study provides certain basis for which found the curcuma antioxidant activity molecules in medicinal plants research,its provides necessary theoretical basis for the curcuma medicinal plants and the scientific application of the genera of medicinal plants. |
