Objective:Proving the ovarian hyperstimulation syndrome will result in the vascular permeability increasing which caused intestinal mucosal damages or not.Can I-FABP as the marker of intestinal mucosa injury in ovarian hyperstimulation syndrome?It will lay the foundation for diagnosis and treatment of intestinal mucosal damages in ovarian hyperstimulation syndrome.Methods:20 female 22-day-old Wistar rats were randomly divided into model group and the control group,each group of 10.In Model Group rats treated with PMSG and hCG,establishing OHSS model?In Normal Group rats treated with the same amount of saline solution.ELISA tests the concentration of I-FABP in serum and the method of Evans blue measures intestinal wall vascular permeability.HE staining observes the change of small intestine tissue morphology of rats,and assesses The degree of damages of intestinal tissue by Chiu's score.Western blot and Immunohistochemistry detects the expression of Occludin and I-FABP protein in small intestine tissues.The Pearson correlation coefficient analysizes the Evans blue leakage quantity,serum I-FABP concentration and the correlation of small intestinal injury Chiu's score.Results:(1)Pathomorphology change and The Chiu's score:In OHSS rats,under the top of the small intestinal villus gap widened,part of the villi falled off,lamina propria bared and inflammatory cell infiltrated.In Normal rats,small intestinal villus arraying in alignment,the structure of blood vessels and serous layer is normal.The Chiu's score of the OHSS rats is significantly higher than the normal rats(3.7 vs 0.2 points,P<0.01).(2)The concentration of I-FABP in serum:The concentration of I-FABP in serum of the OHSS rats is higher than the normal rats(11.612±0.827 vs 1.266±0.132 ng/L,P<0.01).(3)Evans blue leakage quantity:Evans blue leakage quantity of small intestine tissue in Normal rats is significantly lower than OHSS rats(11.04±1.41ug/g vs 33.01±2.63ug/g,P<0.01).(4)The expression of I-FABP and Occludin protein of small intestine:The expression of I-FABP protein is mainly in the area at the top of the small intestinal villus epithelial cell cytoplasm in Immunohistochemistry.The Occludin protein expresses in the area of small intestinal villus epithelial cell membrane.The expression of Occludin and I-FABP protein of small intestine of the OHSS rats is lower than normal rats(P<0.05).(5)Correlation analysis:The Evans blue leakage quantity and I-FABP concentration of serum has significant correlation with the Chiu's score of small intestinal injury.Conclusion:1.The ovarian hyperstimulation syndrome result in the vascular permeability increasing which caused intestinal mucosal damages.2.Serum I-FABP may be regarded as a noninvasive diagnostic biomarkers of small intestinal mucosa injury in OHSS.
