Background and purpose:Heart failure(HF)is a disease characterized by systolic or diastolic dysfunction.The morbidity and mortality of the disease is still high and it is difficult to control the disease effectively.Growth Arrest Specific Protein 6(Gas6)is a vitamin K-dependent protein.Gas6 can regulate the proliferation,migration and apoptosis of endothelial cells and it also regulates vascular morphology.However,the relationship between Gas6 and heart failure is stillunclear.Method:This study is divided into two parts:basic research and clinical research.Basic research part:Treat and incubate the cardiomyocytes(24 and 48hours)of the rats(neonatal rats)with Ang?(10-7 mol/L)in order to construct the cell model of cardiomyocyte hypertrophy,measure the expression of Gas6 in cardiomyocyte and the cell cross-sectional area and preliminary explore the role of Gas6 in heart failure.Clinical research part:This subject is a single-center retrospective study.We brought into 223 cases of heart failure(81 cases)and non-heart failure(142cases)patients which were commited and treated in the Cardiology Division of Shanghai 10th People's Hospital from December 2015 to March 2017.We collected and centrifuged Blood samples after their admission in hospital on the second day,and we measured the plasma concentration of Gas6.In the meanwhile,we collected the patients' basic clinical information,the results of blood tests and cardiac ultrasonography and so on.At last analysed the statistical data.Results:basic research part:Ang? can induce hypertrophy of cardiomyocytes,and the cell area increases with the increasing of treatment time.The content of Gas6 in Ang? treated group was significantly higher than that in control group,and the content of Gas6 in cardiomyocytes treated for 48h was significantly higher than that in 24h treated group.Clinical research part:Of all the selected 223 patients,there were 139 male patients(62.3%)and 84 female patients(37.7%).The average age was 64.3 ± 12.4 years old.The number of patients with heart failure was 81(36.3%)and the number of patients with non heart failure was 142(63.7%).The plasma concentration of Gas6 has a linear relationship with NT-proBNP,left ventricular ejection fraction,C-reactive protein,serum creatinine and D-dimer.The plasma Gas6 concentration in patients with heart failure was significantly higher than that in patients with non heart failure.The worse the heart function was,the higher plasma concentration of Gas6 was.Conclusion:Gas6 is highly expressed in the peripheral blood of patients with HF and myocardial tissue in mice.It is plays an important role in the pathogenesis of HF.Gas6 is closely related to heart function.NT-proBNP and LVEF in patients with heart failure,and may be an important molecular biological marker for evaluating cardiac function.
