[Objective] we established an inflammatory model of the central nervous system,to explore whether Apelin-13 through the PI3K/AKT signaling pathway to improve the central inflammatory response in rats with abnormal central insulin signaling pathway.[Methods] Novel object location test and novel object recognition test were used to measure the learning and memory induced by chronic variate stress co-treated with LPS.The effect of chronic variate stress co-treated with LPS to IL-1? and TNF-? was measured by western blot(WB).The effect of Apelin-13 on chronic variate stress co-treated with LPS-induced increased was measured by WB.The effect of chronic variate stress co-treated with LPS to hippocampal insulin signaling pathway was measured by WB.The effect of Apelin-13 on chronic variate stress co-treated with LPS-induced abnormal of hippocampal insulin signaling pathway was measured by WB.The effect of PI3 K antagonist LY294002 on impairment of learning and memory,abonormal of hippocampal insulin signaling pathway ameliorates by Apelin-13 was measured by WB.[Results] The impairment of learning and memory induced by chronic variate stress co-treated with LPS was ameliorated by Apelin-13.The increased of IL-1? and TNF-? in hippocampus of rats induced by chronic variate stress co-treated with LPS was ameliorated by Apelin-13.The abnormal of hippocampal insulin signaling pathway induced by chronic variate stress co-treated with LPS was ameliorated by Apelin-13.LY294002 blocked the ameliorative effect of Apelin-13 on the impairment of learning and memory,abnormal of hippocampal insulin signaling pathway induced by chronic variate stress co-treated with LPS.[Conclusion] Apelin-13 ameliorates chronic variate stress co-treated with LPS-induced learning and memory impairment through PI3K/Akt pathway mediated central insulin signaling in rats... |
