Background:Endometriosis is a common gynecologic problem in women of reproductive age with a multidimensional etiology,including hereditary,hormonal and immunologic factors.Endometrial cells in endometriosis have similar ability of implant,invasion and distant metastasis as compared with malignant tumor.The exact etiology of endometriosis is still unclarified.Increased adhesion,proliferation and migratory capacity have been detected and involved in the pathophysiology and progression of endometriosis.The complex process of endometrial cells adhesion-invasion-angiogenesis is still a hot research area.Prostaglandins are bioactive lipids that exert an autocrine or paracrine function by binding to specific G-protein-coupled receptors(GPCRs)to activate intracellular signalling and gene transcription.Prostaglandins(PGs)are key regulators of reproductive processes,including ovulation,implantation and menstruation.PGF receptors are widely distributed throughout the body.PGF2a exerts its biological function through interactions with FP receptors.Currently,research on the relationship between PGF2a and endometriosis are very limited.Previous research denmonstranted that there are higher PGF2a levels in peritoneal fluid from patients with endometriosis suggesting that endometriosis is associated with increased PGF2a.This study seeks to investigate the effect of PGF2a in endometrial cells.Wound healing assays and transwell assays were used to confirm the role of PGF2a in migration of endometrial cells.ObjectivesTo investigate the role of PGF2a in the migration of endometrial cells and pathogenesis of endometriosis.MethodWe conducted wound healing assays and transwell assasys of endometrial cells from women with or without endometriosis.Results1.EMs Ectopic and normal endometrial cells treated by 300nm PGF2arevealed higher wound healing capacity when compared with control group(P<0.05).In EMs eutopic endometrial cells,there was no significant woud healing capacity difference between PGF group and control group(P>0.05).2.EMs Ectopic endometrial cells treated by 100nm PGF2? showed increased trans-membrane cell number as compared with control group(P<0.01)while normal endometrial cells treated by 100nm PGF2? showed decreased trans-membrane cell number as compared with control group(P<0.01).ConclusionPGF2a enhanced migration of ectopic endometrial cells,suggesting that it may associated with the pathogenesis and progression of endometriosi.Migration of normal endometrial cells could be inhibited by PGF2a providing a new idea for the diagnosis and treatment of endometriosis.
