| Objectives:In this study,we examined the nural protection of SCI after OECs transplantation and discussed the priority between LP(lumbar puncture)transplantation and traditional orthotopic transplantation which can guide clinical treatment.Methods:OECs were transplanted via LP at level L3–4 1 week after transected SCI at T8.Tissue of spinal from mice were collected at 3d,7d,14d,21d,28d,35d,42d,49d and 56d after LP transplantation and the relative distribution of cells at T8 was quantitated.Injured spine was also tested by immunohistochemistry to assess neuron regeneration and scar bridging 8 weeks post-transplantation.Motor function of mice was evaluated during the observation period,using the Basso Mouse Scale(BMS).Results:OECs were examined and confirmed by cell morphology under phase contrast and Immunofluorescence of NGFR p75.LP transplanted OECs could be detected just 3 days after transplantation(p75+area:0.16 mm2)and accumulated to 0.31 mm2 and0.30 mm2 at 21 days and 56 days post-engraftment respectively.Number of endogenous neurons,-400μm to+400μmfar from the epicenter,in OECs transplanted mice is more than that in SCI mice without engraftment.SCI lesion of mice in control group(164.3±3.97μm)was much longer than that in orthotopic transplantation group(117.2±3.59μm)and LP OECs grafted group(116.7±3.60μm).Grafts of OECs induced significant functional improvements in T8 transection mice just from 3 days after cell injection。Compared with orthotopic transplantation,LP transplantation lead to better recovery in early time.Conclusion:The cell transplantation of OECs can obviously improve the motor function of mice.LP transplantation has better regeneration effects in early time compared with orthotopic transplantation. |
