Objective: MMPs are a group of proteolytic enzymes that are involved in the degradation and remodeling of the extracellular matrix and are a superfamily.TIMPs are inhibitors of MMPs.RSA refers to spontaneous abortion that occurs three times more frequently than three consecutive times.RSA is mostly early abortion,often occurring in the same month of pregnancy.RSA common causes of embryonic chromosomal abnormalities,immune dysfunction,corpus luteum dysfunction,hypothyroidism,etc.,in addition to 50% of patients with unknown etiology.The completion of pregnancy relies on the migration of trophoblast cells to the endometrium,and the ability of trophoblastic invasion in patients with RSA is decreased.MMPs and TIMPs play important roles in cell invasion.This study was designed to evaluate the relationship between the MMP-2 and TIMP-2 in villi and the recurrent spontaneous abortion.Methods: This study selected from March 2013 to December 2013 in our hospital obstetrics and gynecology outpatient treatment of recurrent abortion in 26 patients,aged 24 to 43 years,menopause time 42 to 85 days.Select the same period of treatment of our hospital obstetrics and gynecology clinic termination of pregnancy in 26 cases of normal pregnant women as a normal control group,aged 22 to 41 years,menopause time 42 to 82 days.There was no significant difference in age and menopause between the two groups(P> 0.05).The expression of protein of MMP-2 and TIMP-2 in villi of 26 patients with RSA(RSA group)and 26 normal early pregnant women(control group)were detected by Western blotting,respectively.Results: The expression of MMP-2 protein was significantly lower in villi of the RSA group compared with that of the control group,the difference between RSA group and control group was significant(P < 0.05).Meanwhile,the expression of TIMP-2 protein was significantly higher in villi of the RSA group compared with that of the control group,the difference between RSA group and control group was significant(P < 0.05).Conclusion: There are significant differences between MMP-2 and TIMP-2 expression in villi in RSA patients and normal early pregnant women,indicating that MMP-2 and TIMP-2 may play important role in the recurrent spontaneous abortion.MMP-2 protein expression level was significantly lower than that of the control group,or the expression level of TIMP-2 was significantly higher than that of the control group,or MMP-2 protein expression level was significantly lower than that of the control group and the expression level of TIMP-2 was significantly higher than that of the control group,or the expression of MMP-2 Level and MMP-2 protein expression level imbalance,are likely to occur recurrent miscarriage.
