Objective To investigate the relationship between HPV-16 infection and prostate cancer by using the iPDMS(IntegratedPoly(dimeThysikoxane)based HPV-16 peptide microarray in the seroscreening.Method Antigen sequences from seven proteins of HPV-16 were cut into 20 mer peptides containing overlaps of 10 amino acids,resulting in a total of 241 HPV-16 peptide microarrays.155 serum samples(75prostate cancer subjects and 80 BPH samples)were included in the serological search.Significant analysis of microarray(SAM)was used to identify peptides that performed differential expression between the two groups.Receiver operating characteristic(ROC)analyses were performed to assess the predictive value of the identified peptides when compared with PSA,and a p value < 0.05 was considered statistically significant.Results HPV-16 related antibodies existing in both prostate cancers and BPH patients,specific antibodies-epitope interactions performed different in the two groups.12 peptides were identified as statistically significantly higher responding peptides in prostate cancer samples.Receiver operating characteristic(ROC)analyses showed that the diagnosis model with multi-index combination had higher diagnostic efficiency.The area under the ROC curve was 0.793(95% confidence interval 0.721 to 0.864),with sensitivity of 64.0% while a specificity of 82.5% when set the cutoff value to 2.Area under the ROC curve of PSA was 0.748(95% confidence interval 0.668 to 0.828),with sensitivity of 64% while a specificity of 78.7%.Conclusions1?Results showed HPV-16 related specific antibodies exists in prostate cancer and could be detected by HPV-16 based microarray.2?The changes of specific antibody levels may be closely related to the progression of prostate cancer,their serological tests helps in investigating immune function of prostate cancer patients.3 ? iPDMS based peptide microarray might meet the requirements of in vitro diagnosis,the advantage of miniaturization?high throughput?multiple indicator and lower cost also improves their social and economic value as in vitro diagnostic products.They are worth popularizing in clinical practice.
