Objective: To study the relationship between the pathogenesis of uterine leiomyoma and apoptosis,explore whether or not the apoptosis inhibition involved in the mechanism of uterine leiomyoma developing through comparing uterine leiomyoma the apoptosis state,expression or activation of apoptosis related proteins using 20 samples of uterine leiomyoma tissue and normal uterine muscle tissue adjacent to tumors cell.Methods: 20 cases uterine leiomyoma tissues and normal smooth muscle tissues adjacent to tumors of inpatients from 35 to 56 years old(average age 45±5.45)were collected after hysterectomy operation.All the patients have not been used steroid hormone 3 months before operation.All the specimens were checked by routine pathological examination,diagnosed with uterine leiomyoma after hysterectomy.Grouping: uterine leiomyoma group was called research group,normal uterine smooth muscle tissue was called control group.The DNA of the samples were extracted and the DNA fragmentation analysis were carried out.The expression and activation of apoptosis related proteins were checked and measured by Western Blot,and the expression of Bcl-2,Bax and Bcl-2/Bax ratio,the activation of Caspase-3 and Caspase-9 were analysed in normal smooth muscle tissues adjacent to tumors and uterine leiomyoma tissues.Results: After DNA fragmentation analysis,the DNA broken in normal smooth muscle tissue adjacent to tumors were found with distinct DNA ladder,while at the same time,not obvious DNA ladder can be detected in uterine leiomyoma tissues.In 20 cases samples,research group: DNA ladder were found 5 in 20(25%);control group: DNA ladder were found 5 in 20(75%).Using Western Blot method,compared with control group,the expression of Bax in UL group was obviously less,meanwhile the visible expression of Bcl-2 in research group is significantly higher than control group.In 20 cases samples,the expression of Bcl-2 in research group(0.1020.823 ?)is higher than control group(0.0670.348 ?)while the expression of Bax in research group(0.0730.185 ?)is less than control group(0.0880.429 ?)and the Bcl-2/Bax ratio in research group(0.764.45 ?)is significantly increased compared with the control group(0.370.81?)(P<0.05).Using Western Blot analysis,compared with control group,the activation of Caspase-3 and Caspase-9 increased significantly in research group.Conclusions: The occurrence and development of uterine leiomyoma may be associated with apoptosis inhibition,and this process may involve the expression of apoptosis related proteins Bcl-2 and Bax,Bc-2/Bax ratio change and activation of Caspase-3 and Caspase-9,suggesting that pathogenesis mechanism of uterine leiomyoma may associated with Bcl-2 family and Caspase family.
